摘要
目的:研究大蒜素对乙酸型胃溃疡大鼠模型的防治作用,探讨其抗氧化应激和抑制COX-2信号通路相关机制。方法:胃部乙酸注射法复制大鼠胃溃疡动物模型,同时给予30mg/kg、15mg/kg、7.5mg/kg大蒜素进行干预。连续用药14d后,处死大鼠取胃液测定胃酸浓度、胃蛋白酶含量;取大鼠血清采用生化法检测SOD、MDA、GSH-Px及T-AOC变化,采用ELISA法测定血清胃泌素(gastrin,GAS)及PGE2含量;取大鼠溃疡组织采用RT-PCR和Western-blot法检测COX-1、COX-2、TNF-α及IL-1等因子m RNA和蛋白表达。结果:与胃溃疡模型组大鼠比较,大蒜素30mg/kg可显著降低胃溃疡大鼠胃液胃酸浓度、胃蛋白酶含量,减少血清MDA、胃泌素含量,升高溃疡大鼠血清SOD、GSH-Px、T-AOC及PGE_2水平,下调胃溃疡大鼠胃组织COX-2、TNF-α及IL-1因子mR NA和蛋白表达。结论:大蒜素对乙酸诱发的胃溃疡大鼠模型具有防治作用,其机制可能与抗氧化应激和抑制COX-2信号通路有关。
Objective: To observe the protective effects of allicin in rats with gastric ulcer induced by glacial acetic acid, and to explore its mecha-nisms of anti-oxidative stress and COX-2 signal pathway. Methods: The gastric ulcer rats model were established by the glacial acetic acid injected into stomach tissue. At the same time, the rats were treated with allicln (30mg/kg,15mg/kg,7.5mg/kg) by intragastric administra- tion ( i. g. ), once a day for 14 days. The acidity and pepsin activity of rats" gastric juices were detected. Levels of gastrin, PGE2, SOD, MDA, GSH-Px and T-AOC in rats'serums were detected. Expressions of COX-1, COX-2, TNF-α and IL-1 mRNA and protein in ulcer tissue of rats were analyzed by RT-PCR and western-blot method. Results: Compared with the gastric ulcer model group, the allicin (30mg/kg) decreased the acidity and pepsin activity of gastric juices of gastric ulcer rats, reduced the serum contents of MDA and gastrin, elevated the serum levels of SOD, GSH-Px, T-AOC and PGE2, and down-regulated mRNA and protein expressions of COX-1 , COX-2, TNF-α and IL-1 in the gastric ulcer rats'ulcer tissue. Conclusion: Allicin has the protective effects on gastric ulcer rats induced by glacial acetic acid, and its "mechanism was related to anti-oxidative stress and inhibition COX-2 signal pathway.
