Effects of different extraction methods on extraction rate of Auricularia polytricha polysaccharide

摘要 目的优化不同提取方法对木耳多糖的提取工艺并进行方法比较。方法通过正交实验获得热水浸提法、超声波提取法、微波提取法和高温高压提取法对木耳多糖提取的最佳工艺,并在得率、功效、成本、实用性方面进行比较。结果热水浸提法的最优提取条件为:粒度为100~160目,温度为100℃,料液比为1:20,pH调至5,浸提4 h;超声波提取法的最优提取条件为:粒度为160~300目,料液比1:40,功率400 W,提取50 min;微波提取法的最优提取条件为:木耳粉体粒度为160~300目,料液比1:20,功率700 W,提取时间为50 min;高温高压提取法的最优提取条件为:木耳粉体粒度为100~160目,120℃,1:40料液比,提取80min。结论高温高压法提取率高,其工艺路线简捷,易于工业化生产,是一种木耳综合加工利用的有效方法。 Objective To optimize and compare different extraction techniques of Auricularia polytricha polysaccharides extraction.Methods The optimum extraction conditions of techniques including hot water extraction,ultrasonic extraction,microwave extraction,and high temperature and high pressure extraction of polysaccharides were obtained by orthogonal experiments,and the results were compared in aspects of yield,efficacy,cost and practicability.Results The optimum extraction conditions of hot water extraction were as follows:the particle size was 100~160 mesh,the temperature was 100 ℃,the ratio of material to liquid was 1∶20,pH was adjusted to 5,and the extraction time was 4 h.The optimum extraction conditions of ultrasonic extraction were as follows:the particle size was 160~300 mesh,the ratio of material to liquid was 1∶40,the power was 400 W,and the extraction time was 50 min.The optimum extraction conditions of microwave extraction were as follows:the particle size was 160~300 mesh,the ratio of material to liquid was 1∶20,the power was 700 W,and the extraction time was 50 min.The optimum extraction conditions of high temperature and high pressure extraction were as follows:the particle size was 100~160 mesh,temperature was 80 ℃,the ratio of material to liquid was 1∶40,and the extraction time was 80 min.Conclusion High temperature and high pressure extraction method has high extraction rate,and its process route is simple and easy to realize industrial production,which is an effective method for comprehensive processing and utilization of Auricularia polytricha.
