摘要
目的研究使用纤维素酶辅助提取疏毛罗勒挥发油的工艺,并测定挥发油的抗氧化性。方法采用单因素试验和正交试验相结合的办法研究酶浓度、预处理温度、预处理时间对疏毛罗勒挥发油得率的影响,采用紫外分光光度法测定疏毛罗勒挥发油对羟自由基的清除率。结果最佳提取工艺组合为酶浓度2 mg·m L^(-1)、预处理温度45℃、预处理时间4 h。用最佳工艺进行预处理,疏毛罗勒挥发油得率可达0.62%,比温水浸泡预处理法和直接蒸馏法分别提高了16.98%和58.97%。疏毛罗勒挥发油具有较强的清除羟自由基活性的作用,EC_(50)=4.846mg·m L^(-1)。结论该工艺可用于疏毛罗勒挥发油辅助提取,疏毛罗勒挥发油具有较强的抗氧化性。
Objective To study assisted extraction by cellulase of volatile oil in Ocimum basilicum L and its antioxygenation. Methods Single factor test and orthogonal test were used to study effects of enzyme concentration, pretreatment temperature and pretreatment time on the yield of volatile oil from Ocimum basilicum L. UV spectrophotometry was used to determine the scavenging rate of hydroxyl radical in volatile oil from Ocimum basilicum L. Results The optimum extraction process was that enzyme concentration was 2 mg· mL^-1, pretreatment temperature was 45℃, and pretreatment time was 4h. Under the optimum pretreatment conditions, the yield of Ocimum basilicum Lvolatile oil was 0.62%. Compared with warm water soaking pretreatment and direct distillation method, the yield was increased by 16. 98% and 58. 97% , respectively. The volatile oil of Ocimum basilicum L had a stronger role of scavenging hydroxyl radical, EC50 = 4. 846 mg· mL^-1. Conclusion This process could be used for the assisted extraction of volatile oil from Ocimum basilicum L, and the volatile oil of Ocimum basilicum L has stronger antioxidant activity.
