摘要
为提高白灵侧耳菌株、杏鲍菇菌株及其杂交菌株筛选效率及鉴定的准确性,利用ITS-RFLP标记辅助单核杂交技术对白灵侧耳与杏鲍菇的杂交菌株进行了筛选.对5份白灵菇菌株和4份杏鲍菇菌株进行了种间配对杂交,根据亲本菌株交配型将其分成7个组合进行杂交.结果表明:9份亲本材料共具有6个A因子和7个B因子.利用该技术对杂交处理后的菌株样品进行筛选,获得了3份杂交菌株11D、53D、70D,其中杂交菌株11D与53D的菌丝生长速度与亲本菌株相比较差异显著;杂交菌株70D的菌丝生长速度与亲本菌株XB130DB相比较差异显著,而与亲本菌株BL130DB在菌丝生长速度上差异不显著.
Using ITS-RFLP markers assisted monokaryogenesis hybrid method for screening test the hybrid strains of P. eryngii var. tuoliensis C. J. Mou strains and Pleurotus eryngii. This method can improve efficiency and accuracy for screening the hybrid strains. This test intraspecific hybrid between five P. eryngii var. tuoliensis C. J. Mou strains and four Pleurotus eryngii strains, and divided seven hybrid combinations according to the parent strain mating type . The results show that nine parent strains have six factor A and seven factor B,hybrid samples for this method testing has three hybrid strains.. 11D, 53D and 70D. Hybrid strains llD and 53D has significant difference on hypha growth rate which compared with the parent strains. The hybrid strains 70D has only significant difference on hypha growth rate which compared with the parental strain XB130DB, but no significant difference compared with the parental strain BL130DB.
