葛根素对缺氧性肺动脉高压大鼠的保护作用 被引量:2

Protective Effects of Puerarin on Rats with Hypoxic Pulmonary Arterial Hypertension

摘要 目的:研究葛根素对缺氧性肺动脉高压的治疗作用及作用机制。方法:采用常压低氧培养箱建立大鼠的缺氧性肺动脉高压模型;60只清洁级SD大鼠,随机分成正常对照组、模型组、葛根素组,各组造模前30 min进行腹腔注射,葛根素组注射葛根素注射液20 mg/kg,正常对照组、模型组注射等体积0.9%Na Cl溶液。正常对照组于实验室正常环境下培养,模型组、葛根素组于缺氧环境下培养3 d、7 d、14 d、21 d后进行检测。测定包括肺动脉平均压力(mPAP)、右心室肥厚指数(RVHI)及肺组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果:在常压低氧培养箱中进行大鼠的缺氧性肺动脉高压模型成功建立;模型组mPAP、RVHI、MDA含量显著高于正常对照组,SOD活性明显低于正常对照组(P<0.05,P<0.01)。与模型组比较,14 d、21 d时葛根素组mPAP、RVHI、MDA含量明显降低,SOD活性明显升高(P<0.05,P<0.01)。结论:葛根素能有效抑制缺氧导致的肺动脉平均压力增加及氧自由基增加,减轻其肺血管重构的程度,对缺氧性肺动脉高压具有一定的防治作用。 Objective: To study the therapeutic effects of puerarin on hypoxic pulmonary arterial hypertension and its possible mechanism. Methods: Hypoxic rat model was established in hypoxia incubator with ordinary pressure. 60 SD rats of clean grade were randomly divided into the normal control group, model group and puerarin group. 30 minutes before modeling, each group was received the intraperitoneal injection. The puerarin group was treated with puerarin injection at the dose of 20 mg/kg, and the normal control group and model group were treated with normal saline at equivalent volume. The rats in the normal group were raised in the normal environment, and the other rats were raised in the hypoxic environment. After 3,7,14, 21 days, the mean pulmonary artery pressure (mPAP) , right ventricular hypertrophy index (RVHI), the activity of superoxide dismutase(SOD) and the content of malondialdehyde (MDA) in the lung tissue were detected. Results: Hypoxic rat model was established in hypoxia incubator with ordinary pressure. Compared with the normal control group, the mPAP, RVHI and the content of MDA were significantly increased in the model group, and the activity of SOD was obviously decreased ( P 〈 0.05, P 〈0.01 ). Compared with the model group,after treatment for 14 and 21 days,the mPAP,RVHI and the content of MDA were obviously decreased in the puerarin group, and the activity of SOD was obviously increased ( P 〈 0.05, P 〈 0.01 ). Conclusion : Puerarin can effectively inhibit the increase of mean pulmonary artery pressure and oxygen free radical caused by hypoxia, reduce the degree of pulmonary vascular remodeling, which shows a certain protective effects on hypoxic pulmonary arterial hypertension.
