摘要
目的:观察自然衰老大鼠血清皮质酮(CORT)含量和海马组织超微结构的变化及补肾方左归丸和右归丸的作用,探索补肾方延缓衰老的作用机制。方法:SPF级雄性自然衰老SD大鼠45只,随机分为老年组、左归丸组和右归丸组,每组15只;另设青年组15只,共4组。青年组大鼠常规喂养至5月龄,老年组大鼠常规喂养至24月龄,普通饮食、饮水;从20月龄开始,左归丸组和右归丸组大鼠均按6.75 g/(kg·d)生药量食用饲料,连续给药4个月,每周停药2天。采用ELISA法检测各组大鼠血清CORT含量;透射电镜观察海马齿状回神经细胞超微结构。结果:与青年组比较,老年组大鼠血清CORT的含量显著升高,大鼠海马齿状回神经细胞线粒体参数体密度(Vv)、比表面(δ)、比膜面(δm)和神经细胞突触小泡Vv及突触内线粒体Vv显著降低,差异均有统计学意义(P<0.05)。与老年组比较,左归丸组大鼠血清CORT含量显著降低,左归丸组和右归丸组大鼠海马齿状回神经细胞线粒体Vv、δ、δm和海马神经细胞突触小泡Vv及突触内线粒体Vv显著升高,差异均有统计学意义(P<0.05)。结论:自然衰老大鼠血清CORT含量增高,海马神经细胞出现能量代谢的结构改变,补肾方左归丸和右归丸均可通过降低自然衰老大鼠血清CORT含量、改善其海马神经细胞能量代谢的结构变化而延缓衰老。
Objective:To observe the change of content of serum cortisol(CORT) and ultrastructure of hippocampus,and the effect of Bushen formula Zuogui pill and Yougui pill,then to explore mechanism of delaying senility of Bushen formula.Methods:Selected 45 male SD rats in SPF grade,and divided them into the old group,Zuogui pill group and Yougui pill group with 15 cases in each. Another 15 rats were selected as the young group. There were four groups in total. The rats of the young group and the rats of the old group were given routine feeding, regular diet and drinking till 5-month-old and24-month-old respectively. From 20-month-old,rats of the Zuogui pill group and Yougui pill group were all given feed stuff that contained crude drug of 6.75 g/(kg·d). Medication lasted for four months,and the withdrawal period lasted for two days per week. Detected content of serum CORT in all the groups with enzyme-linked immunosorbent assay(ELISA) method,observed nerve cell ultrastructure of hippocampal dentate gyrus with transmission electron microscope(TEM). Results :Compared with the young group,content of serum CORT in the old group was increased significantly,the volume density of mitochondria(Vv) of nerve cell of hippocampal dentate gyrus,specific surface area(δ),specific membrane(Sm),and neuron synaptic vesicle Vv and its mitochondria Vv were decreased significantly,the differences being significant(P〈0.05). Compared with the old group,content of serum CORT in the Zuogui group was decreased significantly,Vv of nerve cell of hippocampal dentate gyrus,δ and δm,and neuron synaptic vesicle Vv and its mitochondria Vv in zuogui pill group and yougui pill group were increased significantly,the difference being significant(P〈0.05). Conclusion:Serum CORT content of natural aging rats was increased,and structural change occurred in energy metabolism of hippocampal neurons. Bushen formula Zuogui pill and Yougui pill can delay aging by reducing the content of serum CORT of natural aging rats,and improving its structural change of energy metabolism.
