摘要
目的:制备并鉴定对乙酰氨基酚多克隆抗体。方法:以对乙酰氨基酚的特征部分为基础设计合成半抗原丁酸酰氨基酚(PCTM-H),并通过活泼酯法将其与载体蛋白BSA、OVA分别偶联制备免疫抗原PCTM-H-BSA和包被抗原PCTM-H-OVA,再经过动物免疫获得多克隆抗体。结果采用间接竞争ELISA法建立标准曲线,测得其IC50为36.0 ng/m L,定量检测线性范围为2.62~320.54 ng/m L,与其它结构类似物的无交叉反应,添加回收实验其回收率在70.0%~90%之间。结论:成功获得抗对乙酰氨基酚多克隆抗体,该抗体可实现凉茶中非法添加对乙酰氨基酚的快速检测。
Objective: Preparation and identification of polyclonal antibody against paracetamol(PCTM). Methods: The hapten PCTM-H was designed and synthesized on the basis of characteristics part of PCTM and artificial antigens PCTM-H -BSA(immmue antigen)and PCTM-H -OVA(coating antigen) were prepared by coupling with the carrier proteins using active ester method. The polyclonal antibody (PAb) against PCTM was produced by animal immunity. Results Indirect ELISA experiments showed that the IC50 was 36.0 ng/mL, linearity range was from 2.62 to 320.54 ng/mL. There is no cross reactivity against other structure analogous and the recovery is between 70.0 % and 90 % . Conclusion: Successfully obtained the PAb against PCTM, that could detect the illegal additive paracetamol in herbal tea.
