摘要
目的研究红细胞血型系统RhD抗原的蛋白质结构,预测该抗原的T-B细胞联合表位,准确定位T-B联合表位在抗原表面的分布情况。方法采用各种预测软件和在线预测工具,根据RhD抗原的理化性质预测其蛋白质二级结构以及T、B细胞表位,获得T-B联合表位所在的氨基酸残基的位置。结果红细胞RhD抗原有T-B联合表位2个,氨基酸残基所在的位置:36-54,66-82。结论经过生物信息学方法计算获得的2个表位可以作为确定人类红细胞RhD抗原潜在T-B联合优势表位的参考。
Objective To study the protein structure of RhD antigen of erythrocyte blood group system, to predict the T-B cell epitope of this antigen, and to accurately locate the distribution of T-B epitope on anti-gen surface. Methods A variety of prediction software and an online prediction tool were used to predict the protein two structure and the T and B cell epitopes according to the physicochemical properties of the RhD antigen, and to achieve the location of the amino acid residues at the T-B epitope. Results The eryth-rocyte RhD antigen has 2 potential T-B epitopes which the amino acid residues are located at 36 - 54 and 66 -82. Conclusion The two epitopes obtained by bioinformatics methods can serve as a reference for deter-mining the potential T-B joint advantage epitope of human erythrocyte RhD antigen.
