摘要
目的建立小鼠变应性鼻炎(allergic rhinitis,AR)模型,观察金银花提取物对变应性鼻炎小鼠炎性因子水平的影响。方法选取60只健康成年雄性C57BL/6小鼠,按照随机数字法随机分为4组:空白对照组、模型对照组、金银花治疗组、阳性对照组,每组15只。采用卵清蛋白(OVA)建立小鼠AR模型。采取积分法记录AR症状严重程度;取小鼠鼻中隔黏膜,用流式细胞仪分析IL-17来源;ELISA法检测各组小鼠血清中IL-2、IL-4、IL-17、IFN-γ、sIgE的表达水平;小鼠鼻中隔黏膜病理切片计数嗜酸性粒细胞;RT-PCR法检测鼻中隔黏膜中IL-17mRNA的表达;Western blot法检测IL-17蛋白表达水平。结果金银花治疗组及阳性对照组小鼠挠鼻、喷嚏及流涕情况较模型对照组有明显减轻。通过流式细胞仪检测发现IL-17主要来源于γδT细胞。金银花治疗组及阳性对照组小鼠鼻黏膜病理切片嗜酸性粒细胞计数,血清IL-4、IL-17、sIgE水平,组织IL-17mRNA的表达,IL-17蛋白表达水平等,均较模型对照组降低(均P<0.01),而血清IL-2、IFN-γ水平均上升(均P<0.01)。结论金银花提取物可调控OVA致敏的AR小鼠炎性因子的表达,对AR具有免疫调节作用。
Objective To establish a mouse model of allergic rhinitis(AR),and to observe effect of Honeysuckle extract on inflammatory factors in AR mice.Methods Sixty healthy male C57BL/6mice were randomly divided into 4groups:blank control group,model control group,treatment group and control group.AR mouse model was established by using ovalbumin(OVA).AR symptom severity was graded by score.Nasal mucosa of the mice was harvested.IL-17 source was analyzed by flow cytometry.Expression levels of IL-2,IL-4,IL-17,IFN-γand sIgE in serum were detected by ELISA.Eosinophilic granulocytes were counted in nasal mucosa section of the mice.Expression of IL-17 mRNA was detected by RT-PCR.Western blotting was used to detect the expression level of IL-17 protein.Results Symptoms of scratching nose,sneezing and runny nose were significantly alleviated in the Honeysuckle treatment group and positive control group as compared with the control group.The flow cytometry results showed that IL-17 was mainly derived fromγδT cells.Eosinophilic granulocyte count,serum IL-4,IL-7and sIgE,tissue IL-17 mRNA and protein were significantly decreased,while IL-2and IFN-γsignificantly increased in the Honeysuckle treatment group and positive control group as compared with the control group(all P〈0.01).Conclusion The extract of Honeysuckle extract can regulate expression of inflammatory factors in OVA sensitized AR mice,and exert immunomodulatory effect on AR.
