摘要
目的观察不同浓度白藜芦醇及不同作用时间对SH-SY5Y人神经母细胞瘤细胞生长的影响,并对其增殖进行初步研究。方法用MTT方法观察不同浓度(12.5、25、50、100)μmol/L白藜芦醇作用20h、30h、40h后SY5Y细胞存活的数量。用增殖仪器观察不同浓度(32、63、125)μmol/L白藜芦醇作用40h内SH-SY5Y细胞数量变化情况。用显微镜观察不同浓度药物作用20h、30h、40h时的细胞状态。结果 MTT结果显示白藜芦醇对SY5Y细胞的抑制率随着药物浓度的增加而增加,并且每个浓度的抑制率在作用40h最大,P值小于0.05,具有统计学意义。增殖仪器显示白藜芦醇能够抑制该细胞的增殖,并且随着白藜芦醇浓度的增加,抑制细胞增殖的作用越明显。药物作用细胞后显微镜观察可看出,白藜芦醇作用的细胞密度变小,变圆、变亮。随着白藜芦醇浓度的增加,抑制细胞增殖的作用越明显。结论白藜芦醇对SH-SY5Y人神经母细胞瘤细胞的增殖具有抑制作用。
Objectives To observe the effect of resveratrol and different time on the growth of SH-SY5Y human neuroblasto- ma ceils, and to study its proliferation. Methods MTT method was used to observe the survival of SY5Y ceils at different concentrations {12.5,25,50,100} μmol / L resveratrol for 20 h,30 h and 40 h. The number of SH-SY5Y cells in 40 h after differ- ent concentrations resveratrol was observed by proliferative instrument. The cell status was observed by microscope at 20 h,30 h and 40 h. Results MTT results showed that the inhibition rate of resveratrol on SYSY cells increased with the increase of drug concentration, and the inhibition rate of each concentration was the largest at 40 hours, and the P value was less than 0. 05, which was statistically significant. The proliferation apparatus showed that resveratrol inhibited the proliferation of the cells, and with the increase of resveratrol concentration, the effect of inhibiting cell proliferation was more obvious. Effects of drugs on cells could be seen through microscope,and the density of processed with resveratrol becomes smaller, round,bright. With the increase of resveratrol concentration,the effect of inhibiting cell proliferation is more obvious. Conclusion Resveratrol inhibits the proliferation of SH-SYSY human neuroblastoma cells.
