摘要
目的观察喜炎平注射液对腺病毒感染Hep-2细胞Bcl-2/Bax蛋白表达的影响,探讨喜炎平注射液体外抗病毒机制。方法用腺病毒感染Hep-2细胞,分别予喜炎平高、中、低剂量组及利巴韦林干预,采用免疫组织化学法检测各组Hep-2细胞的Bcl-2、Bax蛋白表达情况。结果模型组Bax蛋白表达显著高于正常组,差异有统计学意义(P<0.05);喜炎平各剂量组及利巴韦林对照组Bax蛋白表达均低于模型组,其中喜炎平高剂量组、利巴韦林对照组Bax蛋白表达显著低于模型组,差异有统计学意义(P<0.05)。各组Bcl-2蛋白表达比较差异均无统计学意义(P>0.05)。结论喜炎平注射液通过抑制Bax蛋白表达的上调发挥体外抗腺病毒的作用,高剂量组优于中低剂量组。
Objective To explore mechanism of antiviral mechanism of Xiyanping injection in vitro by invstigating influence of Xiyanping injection on expression of Bcl-2/Bax protein in adenovirus-infected Hep-2 cells. Methods The Hep-2 cells were infected with adenovirus, which were interfered with high, medium and low dose of Xiyanping injection and ribavirin. Then expressions of Bcl-2/Bax protein were measured by immunohistochemistry. Results The expressions of Bax protein in model group were significantly higher than normal groups(P〈0.05). The expression of Bax protein in all Xiyangping groups and ribavirin control group was lower than that of model group, and it was much lower in the high-dose Xiyanping group and ribavirin control group than in the model group(P〈0. 05). The expressions of Bal-2 protein in different groups had no significant difference(P〈0.05). Conclusion Xiyanping injection can kill ADV by suppressing the expressions of Bax protein. The high-dose Xiyanping injection is better than others.
