Expression of signal-induced proliferation-associated gene 1, Ezrin and E-cadherin proteins in epithelial ovarian carcinoma and their clinical significances

摘要 目的 探讨信号诱导增殖相关蛋白1(SIPA1)、Ezrin蛋白和上皮钙黏素(E-cad)与卵巢上皮性癌临床病理参数的关系及其相关性.方法 采用免疫组织化学法检测SIPA1、Ezrin和E-cad在卵巢上皮性癌、交界性卵巢上皮性肿瘤、良性卵巢上皮性肿瘤和正常卵巢组织中的表达.结果 SIPA1在卵巢上皮性癌、交界性卵巢上皮性肿瘤、良性卵巢上皮性肿瘤和正常卵巢组织中的阳性率分别为44.2%(23/52)、64.5%(20/31)、93.3%(28/30)、100.0%(15/15),各组间表达差异有统计学意义(χ^2=29.159,P=0.000);Ezrin的阳性率分别为57.7%(30/52)、61.3%(19/31)、90.0%(27/30)、93.3%(14/15),各组间表达差异有统计学意义(χ^2=14.555,P=0.002);E-cad的阳性率分别为23.1%(12/52)、58.1%(18/31)、86.7%(26/30)、0(0/15),各组间表达差异有统计学意义(χ^2=45.731,P=0.000).在上皮性卵巢癌组织中,SIPA1阳性率与肿瘤病理分级有关(χ^2=3.895,P=0.048),与组织学类型、临床分期无相关性(均P〉0.05);Ezrin的阳性率与肿瘤病理分级、组织学类型和临床分期均无相关性(均P〉0.05);E-cad的阳性表达与肿瘤临床分期有关(χ^2=5.983,P=0.014),与组织学类型、组织学分级无相关性(均P〉0.05).在卵巢上皮性癌中,SIPA1、Ezrin的表达和E-cad均呈正相关(r=0.339,P=0.014;r=0.284,P=0.041),SIPA1与Ezrin的表达无相关性(r=0.214,P=0.128).结论 SIPA1、Ezrin和E-cad在卵巢上皮性癌的发生、发展中起重要作用,三者联合检测可能更好地评价卵巢上皮性癌患者的预后. Objective To investigate the expression of signal-induced proliferation-associated gene 1 (SIPA1), Ezrin and E-cadherin (E-cad), and their relationship with clinical patterns in epithelial ovarian carcinoma.Methods Immunohistochemistry was used to detect the expression of SIPA1, Ezrin and E-cad in normal ovarian tissue, benign epithelial ovarian tumor, borderline epithelial ovarian tumor and epithelial ovarian carcinoma,respectively. Results The positive rate of SIPA1 expression was 44.2 % (23/52), 64.5 %(20/31), 93.3 % (28/30) and 100.0 % (15/15) in epithelial ovarian carcinoma, borderline epithelial ovarian tumor, benign epithelial ovarian tumor, and normal ovarian tissue, respectively, and there was a statistical difference (χ2 = 29.159, P= 0.000). The corresponding rates were 57.7 % (30/52), 61.3 % (19/31), 90.0 %(27/30) and 93.3 % (14/15) for the positive rate of Ezrin expression (χ^2= 14.555, P= 0.002), as well as for 23.1 % (12/52), 58.1 % (18/31), 86.7 % (26/30) and 0 (0/15) for the positive rate of E-cad expression, respectively (χ^2= 45.731, P= 0.000). In patients with epithelial ovarian carcinoma, the expression of SIPA1 was correlated with tumor differentiation (χ^2=3.895, P=0.048), but not with histological type and clinical stage (all P〉0.05). The expression of Ezrin was not correlated with histological type, tumor differentiation and clinical stage (all P〉0.05). There was a positive correlation between expression of E-cad and SIPA1, Ezrin in epithelial ovarian carcinoma, respectively (r= 0.339, P= 0.014; r= 0.284, P= 0.041), but no correlation between the expression of SIPA1 and Ezrin (r= 0.214, P= 0.128). Conclusions SIPA1, Ezrin and E-cad play important roles in the occurrence and development of epithelial ovarian carcinoma. They cooperate in the progression and their combined detection can better evaluate the prognosis of epithelial ovarian carcinoma.
