摘要
目的采用Caco-2细胞模型,研究淫羊藿苷(ICA)吸收转运特性。方法成功制备Caco-2细胞模型后,分为空白组、ICA组、氢离子组、无钠离子组、ATP抑制剂组,给予ICA(10μmol/L)3 h后,采用UPLC-MS分析,比较刷状缘侧、基底侧及细胞样本中ICA及淫羊藿次苷Ⅱ(ICS Ⅱ)的含量,观察不同条件对ICA、ICS Ⅱ吸收的影响。结果与ICA组相比,氢离子组、无钠离子组及ATP抑制剂组刷状缘侧ICA含量显著降低(P<0.05),基底侧ICA含量显著升高(P<0.05),而细胞中仅无钠离子组ICA含量显著增高(P<0.05);与ICA组相比,无钠离子组刷状缘侧、基底侧、细胞中ICS Ⅱ含量均无显著变化,氢离子组基底侧、细胞中ICS Ⅱ含量显著升高(P<0.05),ATP抑制剂组刷状缘侧、基底侧ICS Ⅱ含量显著增高(P<0.05)。结论氢离子、钠离子以及ATP对ICA及ICS Ⅱ的吸收转运有显著影响。
Objective To study the absorption and transportation of icariin (ICA) using Caco -2 cell model. Methods The experiments included the control group, the ICA group, the hydrogen ions group, the sodium ion free group and the ATP inhibitor group. ICA (10 umol/1)was administrated for 3 h. The concentrations of ICA and icariside II( ICS II)in the apical side(AP) ,the basolateral side(BL) and the cell were measured by UPLC/ MS. The effects of hydrogen ion, sodium ion, and ATP on the absorption and transportation were observed. Results Compared with ICA group, the contents of ICA were reduced in AP and increased in BL of the hydrogen ions group, the sodium ion free group, and the ATP inhibitor group (P 〈 0.05 ) , and only increased in cells of the sodi- um ion free group (P 〈 0.05). And there were no significant differences on the contents of ICS II in AP, BL, and cells of the sodium ion free group. For the hydrogen ions group,the contents of ICS II were increased in BL and cells (P 〈0.05 ). For the ATP inhibitor group,the contents of ICS II were increased in AP and BL( P 〈 0. 05 ). Conclusion Hydrogen ion, sodium ion and ATP significantly affect the absorption and transportation of ICA and ICS II.
