摘要
目的:运用均匀设计方法筛选中药组分HJJB方改善非酒精性脂肪肝肝脏脂肪代谢的主效应组分及剂量配比,揭示HJJB方防治非酒精性脂肪肝的部分物质基础。方法:采用高脂饮食诱导的大鼠脂肪肝模型(12周),以HJJB方(红景天苷,绞股蓝总苷,姜黄素,白术多糖)为研究对象,运用均匀设计"4因素8水平设计表"分组设计;以大鼠肝组织甘油三酯(TG)、游离脂肪酸(FFA)、磷酸化的乙酰辅酶A羧化酶(p ACCase)、固醇调节元件结合蛋白-1c(SREBP-1c)、碳水化合物元件结合蛋白(CHREBP)含量为筛选指标,分析不靶点的主效应组分及剂量配比。结果:与正常组相比,模型组的肝组织TG、FFA、p ACCase、SREBP-1c、CHREBP含量均显著升高(P<0.01);均匀设计各组显示出不同强度的药效学效应。降低肝组织TG含量的有效组分是姜黄素、白术多糖,最佳配比是姜黄素4.35+白术多糖69.75。绞股蓝总苷和姜黄素对降低肝组织FFA含量有效,最佳配比为绞股蓝总苷3.54+姜黄素21.76。降低肝组织p ACCase、SREBP-1c含量的有效成分为绞股蓝总苷。而红景天苷则对肝组织CHREBP含量有降低作用。结论:HJJB方能通过多途径调节非酒精性脂肪肝肝脏脂肪代谢,不同途径有其相应的主效应组分和最佳剂量配比。
Objective: Unitorm design method was used to screen the main effective components and dosage ratio of HJJB Compound on improving liver fat metabolism of nonalcoholic fatty liver to reveal the material basis of HJJB com- pound for the prevention and control of nonalcoholic fatty liver disease. Methods : Fatty liver disease in SD male rats were induced by high -fat diet for 12 weeks. With the H.IJB compound (Salidroside, gynostemma total glycosides, curcuminand and atractylodes polysaccharides) as the research object, the grouping design was carried out according to the uniform de- sign U8 (85) table and taking the contents of liver TG, FFA ,pACCase , SREBP- l c and CHREBP as screen index. The main effective components and dosage ratio of different target were obtained by multiple regression analysis. Results : Com- pared with normal group~,the liver TG, FFA,pACCase, SREBP - 1 c and CHREBP levels of model group were significant- ly elevated (P 〈 0.01 ). Uniform design groups show different intensity of pharmacodynamics effect. The eftective compo- nent of decreasing liver TG content is curcumin and Atractylodes polysaceharides. The optimal proportion is 4.35 g curcu- rain + 69.75 g atractylodes polysaccharides. The gynostemma total glycosides and curcumin can decrease the liver FFA content and the optimal proportion is 3.54 g gynostemma total glycosides + 21.76 g curcumin. The main effective compo- nents of reducing liver pACCase and SREBP - 1 c content are gynostemma total glycosides. While salidroside can decrease the content of liver CHREBP. Conclusion: H.IJB Compound can regulate liver fat metabolism of nonalcoholic fatty liver disease through many channels. Different channel has its corresponding main effect and the best dose ratio.
