摘要
目的探讨妇血康颗粒对功能失调性子宫出血模型大鼠凝血功能及性激素水平的影响,分析其可能作用机制。方法75只雌性受孕大鼠被随机分为空白对照组、功血模型组、宫血宁组、低剂量妇血康组、高剂量妇血康组5组,每组各15只,除空白对照组外,其余4组大鼠灌胃米非司酮(8.5 mg/kg)与米索前列醇(100μg/kg),制备功能失调性子宫出血模型。造模成功后第1d,对照组、功血模型组灌服生理盐水,宫血宁组给予宫血宁胶囊(0.07 g/kg),低剂量妇血康组给予妇血康颗粒(100 mg/kg),高剂量妇血康组给予妇血康颗粒(400 mg/kg),连续给药7d。比较各组大鼠断尾后出血时间、凝血功能、激素水平等指标。结果药物干预组大鼠小鼠断尾后出血时间、凝血酶原时间(PT)、凝血酶时间(TT)明显短于功血模型组,纤维蛋白原(FIB)明显高于功血模型组(P<0.05),高剂量妇血康组出血时间、PT、TT明显短于宫血宁组,FIB明显高于宫血宁组(P<0.05);药物干预组(宫血宁组、低剂量妇血康组、高剂量妇血康组)血清卵泡刺激素(FSH)、雌激素(E2)、孕酮(P)明显低于功血模型组,黄体生成素(LH)明显高于功血模型组(P<0.05),高剂量妇血康组血清FSH、E2、P水平明显低于宫血宁组,LH明显高于宫血宁组(P<0.05)。结论妇血康颗粒有助于改善功能失调性子宫出血大鼠凝血功能,缩短出血时间,促进卵泡成熟及排卵。
Objective To explore the effect of Fuxuekang granules on coagulation function and sex hormones levels of dysfunctional uterine bleeding(DUB) rats,and analyze the possible mechanism.Methods Seventy-five female pregnant rats were randomly divided into blank control group,DUB model group,Gongxuening group,low-dose Fuxuekang group,and high-dose Fuxuekang group,15 rats in each group.Except for blank control group,the rats in the other four groups were treated by intragastric administration of mifepristone(8.5 mg/kg) and misoprostol(100 μg/kg) to establish DUB model.On the first day after establishing DUB model,the rats in control group and model group were treated by intragastric administration of normal saline,the rats in Gongxuening group were treated by Gongxuening capsules(0.07 g/kg),the rats in low-dose Fuxuekang group were treated by Fuxuekang granules(100 mg/kg),and the rats in highdose Fuxuekang group were treated by Fuxuekang granules(400 mg/kg),the treatment lasted 7 days.The bleeding time,coagulation function,hormones levels after docking in the five groups were compared.Results The bleeding time,prothrombin time(PT),and thrombin time(TT) after docking in medication intervention groups was statistically significantly shorter than that in DUB model group(P 0.05);the level of fibrinogen(FIB) in medication intervention groups were statistically significantly higher than that in DUB model group(P 0.05);the bleeding time,PT,and TT in high-dose Fuxuekang group was statistically significantly shorter than that in Gongxuening group(P〈0.05);the level of FIB in high-dose Fuxuekang group was statistically significantly higher than that in Gongxuening group(P〈0.05);the levels of follicle stimulating hormone(FSH),estrogen(E2),and progesterone(P) in medication intervention groups were statistically significantly lower than those in DUB model group(P〈0.05);the level of luteinizing hormone(LH) in medication intervention groups were statistically significantly higher than that in DUB model group(P〈0.05);the levels of FSH,E2,and P in high-dose Fuxuekang group were statistically significantly lower than those in Gongxuening group(P〈0.05);the level of LH in high-dose Fuxuekang group was statistically significantly higher than that in Gongxuening group(P〈0.05).Conclusion Fuxuekang granules is beneficial to improve coagulation function,shorten bleeding time,and promote follicle maturity and ovulation of DUB rats.
