烟草是中国重要的经济作物之一。盐胁迫对烟草生产造成巨大危害,克隆盐胁迫响应基因,为从分子水平抵御盐胁迫提供基因资源。本研究克隆了Na+/H+反向转运蛋白基因。该基因编码的蛋白为膜蛋白,具有12个跨膜区。进化分析表明,Nt NHX1-1与番茄的Sl-NHX1遗传距离最近,相似性为95%,其次为甜辣椒的Ca-NHX2,相似性为94%。组织特性表达分析表明,NtNHX1-1基因在茎中表达最高、其次为叶、花和根。NaCl处理后NtNHX1-1基因的表达明显上调,表明该基因可能在烟草抵抗盐胁迫中发挥重要功能。通过调控NtNHX1-1基因的表达有望获得耐盐胁迫烟草新品系。
Tobacco is one of the most important economic crops in our country. Salt stress could cause great loss to tobacco production. Since the salt stress response genes provide gene resources for resisting salt stress on the molecular level, Na+/H+antiporter gene, NtNHX1-1, was the cloned from tobacco in this study. The protein encoded by NtNHX1-1 is a membrane protein with 12 transmembrane regions. Evolutionary analysis showed that genetic distance between NtNHX1-1 the and tomato Sl-NHX1 was nearest, the similarity was 95%, followed by the sweet pepper Ca-NHX2, the similarity was 94%. Tissue specific expression showed that NtNHX1-1 was expressed dominantly in stem, followed by leaves, flowers and roots. After Na Cl treatment, the expression of NtNHX1-1 was significantly increased, which indicated that NtNHX1-1 may play an important function in tobacco resistance to salt stress. By regulating the expression of the NtNHX1-1 gene maybe cultivate the new tobacco lines with tolerance to salt stress.
