摘要
目的:通过高通量测序探寻牙周健康状况和民族对口腔微生物多样性产生的影响。方法:采集12名牙周炎患者(5名汉族和7名裕固族)和10名健康者(5名汉族和5名裕固族)的唾液,提取细菌总DNA,用Illumina MiSeq高通量测序平台对16SrRNA基因序列进行测序分析。结果:在UniFrac距离总变化中,牙周因素在未加权分析中有差异(P=0.02),民族因素在加权分析中有差异(P=0.03),两种因素的相互作用在加权和未加权分析中均有差异(P<0.05)。结论:牙周健康状况及民族因素对口腔微生物多样性均有影响作用,并可能存在协同作用,但牙周健康状况是引起细菌组成变化的主要因素。
Objective:To explore the effects influenced by the status of periodontal health and different ethnicity on oral microbial diversity by high throughput sequencing.Methods:Saliva was collected from 12 periodontitis patients(5Han and 7Yugur)and 10 healthy sub-jects(5Han and 5Yugur).The total DNA of bacterias was extracted,then the sequence of 16 SrRNA gene was sequenced by Illumina MiSeq high throughput sequencing platform.Results:In the UniFrac distance total variation observation,the periodontal factors only explained the variation in the unweighted analysis,and the ethnic factors just explained the variation in the weighted analysis.The interaction of the two influencing factors explained the variation in both unweighted and weighted analysis.Conclusion:Periodontal health status and ethnic factors have effects on oral microbial diversity,and there may be synergistic effects,but periodontal health status is the main factor in changing the bacterial composition.
