摘要 建立了一种基于液相的快速测定强化奶粉中β-胡萝卜素方法。样品用无水乙醇-水溶解,添加KOH溶液皂化,皂化完成后水洗,用正己烷与乙酸乙酯混合液萃取,使用流动相溶解后直接用于色谱测定。液相分离柱选取C18柱,通过对色谱分离条件进行优化。结果表明,最佳色谱分离条件确定为以甲醇-二氯甲烷比例为70∶30(V/V)为流动相;流速设置为1.0mL/min;采用PDA检测器,检测波长设为450nm。结果表明,此方法的三水平样品回收率(n=5)为98.50%~100.68%;重复实验结果,显示八次重复性实验的相对标准偏差(RSD)为0.42%;检测限(S/N=3)为5ng。 A method of determination of β-carotene in fortified milk powder by HPLC has been developed. After the samples were dissolved using anhydrous ethyl alcohol,the saponification was induced by adding KOH solution. β-carotene in the milk powder was extracted in hexane-ethyl acetate mix and dissolved in mobile phase solution for HPLC measurement,on the C18 column with the mobile phase composed of methyl alcohol: methylene chloride(70 ∶30,V/V).The flow rate of mobile phase was set at 1.0mL/min. The sample was detected by the PDA detector at 450 nm wavelength. The average recoveries were 98.50%~100.68%. The relative standard deviations(RSD) was 0.42%(n=8)and the detection limi(tS/N=3)was 5.0 ng.
