血管基膜衍生多功能肽抑制肺癌裸鼠移植瘤生长和血管生成 被引量:2

Recombinant vascular basement-membrane-derived multifunctional peptide inhibits the growth and angiogenesis of lung carcinoma in nude mouse xenograft model

摘要 目的:研究重组血管基膜衍生多功能肽(rVBMDMP)对人肺癌裸鼠移植瘤生长和血管生成的抑制作用。方法:人肺癌A549裸鼠移植瘤模型治疗实验,评价rVBMDMP治疗人肺癌的有效性;CD34免疫组化染色检测rVB-MDMP对移植瘤血管生成的影响。结果:rVBMDMP显著抑制人肺癌裸鼠移植瘤的生长,呈剂量依赖性趋势。与模型组相比,5 mg/kg rVBMDMP对人肺癌裸鼠移植瘤的肿瘤生长抑制率是74%;CD34免疫组化染色结果表明5 mg/kgrVBMDMP组微血管密度为(38.56±4.64),显著低于模型组(91.82±6.48)。结论:rVBMDMP对人肺癌裸鼠移植瘤生长和血管生成具有抑制作用。 Objective To investigate the inhibitory effects of recombinant vascular basement-membrane-derived multifunctional peptide(rVBMDMP) on the growth and angiogenesis of human lung carcinoma in nude mouse xenograft model.Methods Human lung cancer in nude xenograft mouse model was employed to evaluate the therapeutic effect of rVBMDMP in vivo.The anti-angiogenic effect of rVBMDMP was detected using immunohistochemical staining for CD34.Results rVBMDMP significantly inhibited tumor growth of human lung carcinoma in nude mouse xenograft model.rVBMDMP(5mg/kg) decreased the tumor volume by 74%,compared with the model group.Immunohistochemical staining for CD34 showed that microvessel density(38.56±4.64) treated with 5mg/kg rVBMDMP was significantly decreased,compared with that(91.82±6.48) in the model group.Conclusion rVBMDMP inhibites the growth and angiogenesis of human lung carcinoma in nude mouse xenograft model.
