依那普利对糖尿病大鼠心肌IGF-1表达的影响 被引量:1

摘要 为观察马来酸依那普利对糖尿病大鼠心肌中胰岛素样生长因子1(IGF-1)表达的影响,探讨马来酸依那普利对糖尿病心肌病可能具有的保护作用及机制。方法把实验大鼠60只适应性喂养2周后随机分为2组,正常(NC)组10只给予普通饲料饲养。待造模组50只,利用腹腔注射链脲佐菌素(STZ)造模,给予高糖高脂饮食,两周后将成模的40只大鼠随机分为2组。糖尿病(DM)组20只继续给予高糖高脂饮食。药物干预(DS)组20只给予马来酸依那普利灌胃10 mg/kg/d,8周后对比3组大鼠以下指标:一般状况(体重、饮食、尿量、毛色、精神状态)、血液生化检查(TC、TG、FBG、Hb A1c水平)、大鼠心肌切片HE染色心肌结构变化、石蜡包埋心肌组织行免疫组织化学染色测定IGF-1含量。结果一般状况:DM组和DS组比NC组体重明显减轻(P<0.05),饮食尿量增多,毛色干枯、精神萎靡不好动。而DM和DS两组间差异无统计学意义。血生化指标:DM和DS组TG、TC、FBG、Hb A1c值较NC组有明显升高(P<0.01),而DM和DS两组间差异无统计学意义(P>0.05)。心肌切片HE染色:DM组较NC组有心肌肥大及心肌纤维化改变趋势,而DS组心肌变化介于DM和DS之间。心肌切片IGF-1免疫组织化学染色:DM组较NC组IGF-1含量增高(P<0.01),而DS组低于DM组(P<0.05)高于NC组(P<0.05)。结论马来酸依那普利可以通过减少糖尿病大鼠心肌中IGF-1因子,抑制糖尿病心肌中心肌细胞的肥大和纤维化趋势。 To observe the effect of maleic acid in the expression of insulin-like growth factor 1(IGF-1) in diabetic rats,and to discuss the protective effect and mechanism of the maleic acid in the diabetic cardiomyopathy.The experimental rats were divided into two groups after two weeks of adaptive feeding, and the normal(NC) group of 10 was raised for regular feed. The 40 rats were randomly divided into two groups in two weeks after the model was made of 50, which was formed by using a peritoneal syringe(STZ), which was given a high-sugar, high-fat diet. The diabetes group(DM) group of 20 continued to give high-sugar and high-fat diets. The drug intervention(DS) group of 20 gave the maleic acid to the stomach 10mg/kg. D, 8 weeks later compared to three groups of rats: General conditions(body weight, diet, urine volume, color, mental state), blood biochemical examination(TC, TG, FBG,Hb A1 c level), the rat myocardial biopsy HE dyed myocardial structure change, paraffin embedding myocardial tissue line immunohistochemical staining content determination of IGF-1.General condition: the DM group and the DS group were significantly less weight than the NC group(P0.05), the amount of urine was increased, the hair color was dry and the spirit was not active. The differences between DM and DS were not statistically significant. Blood biochemical indexes: DM and DS group TG, TC, FBG,Hb A1 c value has significantly increased in the NC group(P0.05). Myocardial biopsy: the DM group was associated with a change in myocardial hypertrophy and myocardial fibrosis in the NC group, and the DS group was between DM and DS.(P0.01), while the DM and DS there was no statistically significant difference between the two groups(P0.05) Myocardial biopsy: the DM group was associated with a change in myocardial hypertrophy and myocardial fibrosis in the NC group, and the DS group was between DM and DS. Myocardial biopsy IGF-1 immunohistochemical staining: the DM group was higher in IGF-1 than the NC group(P0.01),and the DS group was lower than the DM group(P0.05) higher than the NC group(P0.05).The maleic acid innappelle can inhibit the hypertrophy and fibrosis of heart muscle cells in diabetic myocardial cells by reducing the IGF-1 factor in the heart muscle of diabetic rats.
