摘要
目的:探讨膝痹膏摩法对膝痹兔滑膜基质金属蛋白酶的影响。方法:66只健康的新西兰大白兔,随机分10只为正常组对照,剩余用石膏铁丝固定法复制膝痹动物模型,8周时在模型中随机处死6只动物查看造模情况,造模成功后将其随机分模型组、手法组和膝痹膏高浓度、中浓度、低浓度组,膝痹膏3个浓度组给予膝痹膏摩法干预,手法组给予单纯手法,空白组和模型组不干预;干预8周处死动物,取滑膜利用ELISA试剂盒检测MMP1、MMP3、MMP13表达。结果:病理检测观察膝痹膏摩法不同浓度组均可改善关节软骨组织学结构,模型组膝关节软骨出现明显的关节软骨损伤。模型组中MMP-1、MMP-3、MMP-13含量均高于其余5组(P<0.05),正常组中MMP-1、MMP-3、MMP-13含量最低(P<0.05)。各组间比较:膝痹膏中浓度组MMP-1、MMP-3的含量明显低于高浓度、低浓度和手法组(P<0.05),而高浓度、低浓度和手法组之间比较没有统计学意义;膝痹膏中浓度组MMP-13低于低浓度和手法组(P<0.05),但与高浓度比较没有统计学意义。结论:膝痹膏摩法可明显改善膝痹的病理表现,其作用机制可能与3个金属蛋白酶的含量降低有关,进而降低了软骨基质的降解,缓解膝痹的进程,仍需进一步研究其具体的作用靶点,完善其作用机制。
Objective:To investigate the effect of Xibigao method on rabbit’s knee osteoarthritis synovial matrix metalloproteinase.Methods:A total of 66 healthy New Zealand white rabbits were randomly divided 10 as the normal control group and the remaining wire fixation were to set up the plaster copy knee paralysis models.At eighth week,randomly kill 6 model animals to check modeling cases.The successful models were randomly divided into model group,practice group and high,middle and low concentrations of Xibigao groups.Three Xibigao groups were given different concentrations of Xibigao.The practice group was given massage techniques.The blank group and model group were given non-intervention.After 8 weeks,animals were killed using an ELISA kit for the detection of synovial MMP1,MMP3 and MMP13 expressions.Results:Pathological examination of the knee showed different concentrations of Xibigao can improve the histological structure of articular cartilage.The model group had significant articular cartilage damage.Model group’s MMP-1,MMP-3 and MMP-13 were higher than those of the left five groups(P〈0.05).The normal group’s MMP-1,MMP-3 and MMP-13 contents were the lowest(P〈0.05).Comparison between groups:middle concentration of Xibigao group’s MMP-1 and MMP-3 contents were significantly lower than those of high and low concentration of Xibigao groups and massage group(P〈0.05).And the high and low concentration of Xibigao groups and manipulation group did not have statistical significance.Middle concentration of Xibigao group’s MMP-13 was lower than that of the low concentration of Xibigao group and manipulation group(P〈0.05),but was not statistically significant compared with that of high concentration group.Conclusions:Xibigao method can significantly improve knee pathology of paralysis and its mechanism may be related to decreasing three metalloproteinase,and thus reducing the degradation of the cartilage matrix.The knee paralysis mitigation still needs further study of its specific role targets,improving its mechanism of action.
