摘要
目的:探讨芒果苷对链脲佐菌素(STZ)诱导的糖尿病大鼠肾脏损伤的保护作用及相关机理。方法:将SD大鼠分为正常对照组、糖尿病模型组及芒果苷低、中、高剂量组,采用腹腔注射STZ的方法复制糖尿病模型,生化方法检测大鼠血糖、血肌酐、尿蛋白、血和肾脏的SOD活性及MDA含量,ELISA法检测各组血清及肾CTGF的表达,HE染色观察肾脏形态学变化。结果:与模型组比较,芒果苷各组大鼠血糖、血肌酐、24 h尿蛋白量明显下降;血清及肾组织SOD活性升高,MDA含量显著降低,血清及肾脏CTGF蛋白水平与糖尿病模型组相比显著下降。结论:芒果苷能够延缓糖尿病大鼠肾脏损伤,其机制可能与减轻肾脏氧化应激程度及下调肾脏CTGF蛋白的表达有关。
Objective :To investigate the protective effect of mangiferin on diabetic nephropathy and the potential mechanisms in diabetic rat models induced by streptozotocin (STZ).Methods:Healthy male Sprague-Dawley rats were randomly divided into group of normal controls, diabetic models, diabetic models treated with mangiferin in low, middle or high dose.Diabetic rats were induced by intraperitoneal injection with STZ. Biochemistry was used to determine the blood glucose,serum creatinine,urine protein,activities of superoxide dismutase(SOD) and catalase in serum and kidney as well as content of malonaldehyde (MDA) in the kidney, and ELISA was performed to assay the connective tissue growth factor(CTGF) expression in the kidney.The renal tissues were stained with HE and observed by microscopy.Results : Diabetic rats treated with mangiferin had significantly decreased levels of blood glucose,serum creatinine and 24-hour urine protein, yet markedly increased SOD activity in the serum and the kidney and reduced MAD level compared to the sim- ple model rats.The levels of CTGF in renal cortex were dramatically decreased in rats treated with mangiferin compared with diabetic models and normal controls.Conclusion: Mangiferin may delay the kidney injury in diabetic rats.The potential mechanisms may be associated with decreased oxidative stress and down-regulated expression of CTGF protein in renal tissues of diabetic rats induced by STZ.
