摘要
目的:应用去卵巢大鼠建立绝经后骨质疏松(Postmenopausal osteoporosis,PMOP)模型,观察蒙药蓝刺头对去卵巢骨质疏松大鼠骨组织雌激素受体α和β mRNA的影响,为蒙药治疗绝经后骨质疏松提供实验依据。方法:取4mo龄雌性Wistar大鼠84只,按照随机对照原则,将大鼠分为模型组、假手术组、蓝刺头高、中、低剂量组、己烯雌酚组及强骨胶囊组7组,每组12只,假手术组只摘除少量脂肪组织,其余各组均摘除双侧卵巢,造模成功后,正常条件下饲养90d后给予相应药物灌胃,连续灌胃90d后处死大鼠,分离大鼠双侧胫骨,用RTPCR法检测胫骨近端骨组织雌激素受体α和β mRNA表达情况。结果:蒙药蓝刺头能明显改善去卵巢骨质疏松大鼠的一般状况;与模型组相比己烯雌酚组、蒙药蓝刺头各剂量组及强骨胶囊组均提高了ERα、ERβm RNA的表达;与己烯雌酚组相比,蓝刺头中剂量组提高了ERβm RNA的表达;与强骨胶囊组相比蓝刺头各剂量组对ER m RNA表达均无明显差异。结论:蓝刺头能够上调去卵巢大鼠ERα和ERβ的表达,说明蒙药蓝刺头可能通过促进骨形成,调节骨吸收与骨形成的偶联,降低骨转换率,达到防治PMOP的目的。
Objective:Studying the effects of Mongolian echinops on some indexes of ovariectomied rats,the estrogen receptor α and β mRNA to search for the mechanism of drug action.Methods:It selected 84 female Wistar rats which are 4 months old in the study,they were randomly divided into 7 groups:shamed operations group,model group,Qianggu capsule group,diethylstilbestrol group,Mongolian echinops at high、medium、low dose groups.every group had twelve rats.Prepared model reference to the literature,shamed operations group were shamming operated;which removed a little amount of adipose tissue around ovarian.Others were removed bilateral ovarian.Then,it fed rats for 90 days in the normal conditions.And it gived rats corresponding drug orally,intragastrically.It separated rats bilateral tibial bone,and to tissue the estrogen receptor α and β mRNA expression of rats by RT-PCR method.Results:The Mongolian medicine Echinops can improve the general condition of ovariectomized rats significantly.Among the group of diethylstilbestrol,model group and Echinops high group,which improved expression ERα,ERβ mRNA.Echinops dose drug of middle can increase expression of ERβ mRNA than the diethylstilbestrol.There is no difference between the Echinops dose group and strong bone capsule group,the expression of ER mRNA.Conclusion:Echinops can raise expression of ovariectomized rats ERα and ERβ.In short,it is possible to specify Echinops treated osteoporosis.Therefore,Mongolian Echinops may promote bone formation,adjusted the coupling of bone resorption and reduced the rate of bone turn over to reache combating the PMOP.
