颅脑外伤患者血清及脑脊液中Nogo-A基因表达量与神经损伤程度、炎性因子含量的关系 被引量:8

Relationship of Nogo-A gene expression in serum and cerebrospinal fluid with nerve injury and inflammatory factor levels in patients with craniocerebral trauma

摘要 目的:探讨颅脑外伤患者血清及脑脊液中Nogo-A基因表达量与神经损伤程度、炎性因子含量的关系。方法:收集在本院接受治疗的颅脑外伤患者82例作为观察组,同期在本院接受腰麻后内痔手术治疗的正常人58例作为对照组。采用荧光定量PCR法检测两组研究对象血清及脑脊液中Nogo-A基因表达量,采用酶联免疫吸附法(ELISA)测定两组血清中神经损伤指标、炎症因子的含量。采用Pearson检验评估颅脑外伤患者血清及脑脊液中Nogo-A基因表达量与病情严重程度的相关关系。结果:观察组血清及脑脊液中Nogo-A mRNA的表达量高于对照组;血清中IGF-Ⅱ的含量低于对照组,NSE、MBP、S100B的含量高于对照组;血清中HSP-70、PCT、IL-1β、IL-6、CRP的含量高于对照组(P<0.05)。经Pearson检验发现,颅脑外伤患者血清及脑脊液中Nogo-A基因表达量与神经损伤程度、炎性因子含量直接相关。结论:颅脑外伤患者存在Nogo-A基因高表达,且其表达量与患者神经损伤程度、全身炎症反应直接相关。 Objective:To study the relationship of Nogo-A gene expression in serum and cerebrospinal fluid with nerve injury and inflammatory factor levels in patients with craniocerebral trauma.Methods:A total of 82 patients with craniocerebral trauma who were treated in our hospital between January 2015 and February 2017 were collected as observation group,and 58 normal subjects who received internal hemorrhoids surgery under lumbar anesthesia in our hospital during the same period were collected as control group.Fluorescence quantitative PCR method was used to detect Nogo-A gene expression in serum and cerebrospinal fluid of both groups,and enzyme-linked immunosorbent assay(ELISA)was used to determine serum levels of nerve injury indexes and inflammatory factors in two groups.Pearson test was used to assess the relationship of Nogo-A gene expression in serum and cerebrospinal fluid with disease severity in patients with craniocerebral trauma.Results:Nogo-A mRNA expression in the serum and cerebrospinal fluid of observation group were higher than those of the control group;serum IGF-Ⅱlevel was lower than that of control group while NSE,MBP and S100 Blevels were higher than those of control group;serum HSP-70,PCT,IL-1β,IL-6and CRP levels were higher than those of control group(P〈0.05).Pearson test showed that Nogo-A gene expression in serum and cerebrospinal fluid of patients with craniocerebral trauma were directly correlated with the nerve injury degree and inflammatory factor levels.Conclusions:Nogo-A gene is highly expressed in patients with craniocerebral trauma,and its expression is directly correlated with the nerve injury and systemic inflammatory response.
