β-catenin、Smad3及TGF-β1在宫腔粘连患者子宫内膜组织中的表达及意义 被引量:7

Expressions and significance of β-catenin,Smad3 and TGF-β1 in endometrium of patients with intrauterine adhesion

摘要 目的研究β-catenin、Smad3及TGF-β1在宫腔粘连(IUA)患者粘连组织中的表达水平,探讨其在IUA中的作用机制。方法选择2014年5月至2016年5月在乌鲁木齐市妇幼保健院行宫腔镜下宫腔粘连松解术的IUA患者60例为实验组,常规行宫腔镜检查患者60例为对照组。采用实时荧光定量聚合酶链反应(RT-PCR)方法检测实验组IUA患者的粘连组织、靠近粘连的子宫内膜和对照组的正常子宫内膜组织中β-catenin m RNA、Smad3m RNA及TGF-β1 m RNA的表达水平。结果对照组患者正常子宫内膜组织中与实验组轻度、中度、重度IUA患者粘连组织中的β-catenin m RNA、Smad3 m RNA、TGF-β1 m RNA的表达比较,从低到高依次排序:对照组<实验组轻度粘连<实验组中度粘连<实验组重度粘连,对比差异有统计学意义(F值分别为20.35、19.62、22.32,均P<0.05),β-catenin m RNA、Smad3 m RNA、TGF-β1 m RNA在实验组IUA患者粘连组织、靠近粘连的子宫内膜中的表达均高于对照组正常子宫内膜,差异有统计学意义(P值分别为0.038、0.003、0.004);β-catenin m RNA、Smad3m RNA、TGF-β1 m RNA在粘连组织中的表达高于靠近粘连的子宫内膜,差异有统计学意义(P值分别为0.016、0.001、0.008)。在IUA患者粘连组织中TGF-β1与Smad3的表达呈正相关(rs=0.44,P=0.01);TGF-β1与β-catenin的表达亦呈正相关(rs=0.42,P=0.02);Smad3与β-catenin的达无相关性(rs=0.037,P=0.85)。结论在IUA中β-catenin、TGF-β1及Smad3的高表达可能与宫腔粘连的发生有密切相关;β-catenin和TGF-β1/Smad通路可能协同参与子宫内膜纤维化及促进宫腔粘连的形成。 Objective To study the expression levels of β-catenin,Smad3 and TGF-β1 in adhesion tissue in patients with intrauterine adhesion( IUA) and to explore its mechanism in IUA. Methods A total of 60 cases of IUA treated with hysteroscopic lysis of intrauterine adhesions from May 2014 to May 2016 in Urumqi Maternal and Child Health Care Hospital were selected in experimental group,and 60 cases undergoing routine hysteroscopy were selected in control group. Real time fluorescent quantitative polymerase chain reaction( RTPCR) was used to detect the expression levels of β-catenin m RNA,Smad3 m RNA and TGF-β1 m RNA in adhesion tissue and endometrium near adhesion of IUA patients in experimental group and in normal endometrium in control group. Results Sequence of expression levels ofβ-catenin m RNA,Smad3 m RNA and TGF-β1 m RNA in normal endometrium in controls and in adhesion tissues in patients with mild,moderate and severe IUA was controls,mild IUA,moderate IUA and severe IUA patients from low to high,and difference had statistical significance( F value was 20. 35,19. 62,and 22. 32,respectively,all P〈 0. 05). Expressions of β-catenin m RNA,Smad3 m RNA and TGF-β1 m RNA in adhesion tissues and endometrium near adhesions of IUA patients in experimental group were higher than those in normal endometrium in control group,and difference was statistically significant( P value was 0. 038,0. 003 and 0. 004,respectively).Expressions of β-catenin m RNA,Smad3 m RNA and TGF-β1 m RNA in adhesion tissue were higher than those in endometrium near adhesion,and difference was statistically significant( P value was 0. 016,0. 001 and 0. 008,respectively). Expression of TGF-β1 in adhesion tissue of IUA patients was positively correlated with that of Smad3( rs= 0. 44,P = 0. 01). Expression of TGF-β1 was also positively correlated with that of β-catenin( rs= 0. 42,P = 0. 02),but expression of Smad3 had no correlation with that of β-catenin( rs=0. 037,P = 0. 85). Conclusion High expressions of β-catenin,TGF-β1 and Smad3 in IUA patients may be closely related to the occurrence of IUA. Pathway of β-catenin and TGF-β1/Smad may be involved in endometrial fibrosis and promote the formation of IUA.
