肾去交感神经术对阿霉素诱导的心衰大鼠心功能的影响 被引量:1

Effects of renal sympathetic denervation on cardiac function in rats with Adriamycin induced heart failure

摘要 目的 探讨肾去交感神经术(RDN)对阿霉素诱导的心衰大鼠心功能的影响.方法 42只雄性SD大鼠随机分为对照组(n=10,腹腔注射生理盐水)、假手术组(n=16,腹腔注射阿霉素4 mg/kg,每周1次,持续6周制备心衰模型后行假RDN术)和RDN组(n=16,腹腔注射阿霉素4 mg/kg,每周1次,持续6周制备心衰模型后行RDN术).术前及术后4周分别采用超声心动图测定大鼠心功能,ELISA法测定血清BNP水平,蛋白印迹法测定心肌组织血管紧张素(Ang)Ⅱ、胶原蛋白I和金属基质蛋白(MMP)2的表达水平.结果 ①心功能指标LVEF、LVEDd、IVST:与本组术前比较,对照组未见统计学差异(P〉0.05),假手术组和RDN组差异均有统计学意义(P〈0.05).与对照组比较,除假手术组IVST外差异均有统计学意义(P〈0.05);与假手术组比较,术前未见统计学差异(P〉0.05),术后4周除IVST外差异均有统计学意义(P〈0.05).②血清BNP水平:术后4周假手术组和RDN组大鼠血清BNP水平明显高于对照组[(216.14±23.56)pg/ml比(42.72±8.67)pg/ml,(86.17±12.35)pg/ml比(42.72±8.67)pg/ml],RDN组BNP水平明显低于假手术组[(86.17±12.35)pg/ml比(216.14±23.56)pg/ml],差异均具有统计学意义(P〈0.01).③心肌组织AngⅡ、胶原蛋白Ⅰ和MMP2表达水平:与对照组比较,假手术组和RDN组差异均有统计学意义(P〈0.01);与假手术组比较,RDN组差异具有统计学意义(P〈0.01).结论 RDN可改善阿霉素诱导的心衰大鼠心功能,其机制可能与调节AngⅡ、胶原蛋白Ⅰ和MMP2表达水平有关. Objective To investigate the effect of renal sympathetic denervation on cardiac function inrats with adriamycin induced heart failure.Methods 42 male SD rats were randomly divided into three groups:control group (n=10,Intraperitoneal injection of normal saline);Sham operation group (n=16,Intraperitoneal injection of doxorubicin 4 mg/kg,once a week,for 6 weeks to prepare the model of heart failure after sham RDN);RDN group (n=16,Intraperitoneal injection of doxorubicin 4 mg/kg,1 times per week,for 6 weeks to prepare the model of heart failure after RDN).The cardiac function was measured by echocardiography before and 4 weeks after operation,ELISA method for the determination of serum BNP levels.The expression level of angiotensin (Ang) Ⅱ,collagen Ⅰ and metal matrix protein (MMP) 2 was determined by Western blot.Results (1)Heart function index LVEF,LVEDd,IVST:compared with the preoperative group,the control group was not statistically significant (P〉0.05).There were statistically significant differences between the sham operation group and the RDN group (P〈0.05).Compared with the control group,the difference was statistically significant (P〈 0.05) in the sham operation group (IVST).Compared with the sham operation group,there was no significant statistical significance(P〉0.05) before operation.There were statistically significant(P〈0.05) in addition to IVST 4 weeks after surgery.(2)Serum BNP level:The serum levels of BNP in sham operated group and RDN group were significantly higher than those in control group after 4 weeks [(216.14±23.56)pg/ml vs (42.72±8.67)pg/m1,(86.17±12.35)pg/ml vs (42.72±8.67)pg/ml].The level of serum BNP in RDN group was significantly lower than that in sham operation group [(86.17±12.35)pg/ml vs (216.14±23.56)pg/ml].The difference was statistically significant (P〈0.01).(3)The expression level of Ang Ⅱ,collagen Ⅰ and MMP2 in cardiac muscle:Compared with the control group,the difference was statistically significant between sham operation group and RDN group (P〈0.01).Compared with sham operation,the difference of RDN group was statistically significant (P〈0.01).Conclusion RDN can improve cardiac function in rats with adriamycin induced heart failure.The mechanism may be related to the regulation of Ang Ⅱ,collagen Ⅰ and MMP2 expression levels.
