摘要
目的建立薄层色谱和含量测定方法控制黑豆衣质量。方法采用显微鉴别和薄层色谱鉴别对其进行定性;采用高效液相色谱梯度洗脱法对其进行含量测定。结果黑豆衣显微特征明显,具有专属性;薄层鉴别斑点清晰。矢车菊素-3-O-葡萄糖苷在0.05~1.00μg与峰面积呈良好的线性关系(r=0.9998),低、中、高浓度平均回收率分别为97.8%、98.7%、98.9%,RSD分别为0.77%、0.86%、0.15%。结论本方法重复性好、灵敏准确,可用于黑豆衣中主要成分的鉴别和含量测定。
Objective To control the quality of Sojae Semen Nigrum spermoderm by the TLC and content determination method. Methods Microscopical identification and TLC were used for qualitative determination, and HPLC method was used to determine the contents. Results The micro-character was obvious and specific; the spots were clear and accurate. Cyanidin-3-O-glucoside showed a good linearity with peak area (r = 0.9998) at 0.05 - 1.00 μg. The low, medium and high concertration of average recoveries was 97.8%, 98.7%, and 98.9% with RSDs 〈 1.0%. Conclusion The method is accurate, sensitive and reproducible, which can be used to identify and determine the content of Soiae Semen Nigrum spermoderm.
