一株短蛸消化道产蛋白酶菌株的分离鉴定及其酶学性质研究

摘要 用透明圈法从健康短蛸肠道中分离出一株产蛋白酶海洋细菌S-8。通过生理生化实验、16s rDNA序列比对和扫描电子显微镜观察对菌株进行菌种鉴定,并对其产酶进行酶学性质研究。结果表明:菌株S-8为灿烂弧菌,命名为灿烂弧菌S-8。菌株S-8产蛋白酶活性良好,其粗酶液蛋白酶活性为89.12U/mL,SDSPAGE电泳表明菌株S-8所产蛋白酶分子量分别为149.6k D、127.4KD、78.9KD、61.7KD、57.3KD、40.3KD。酶学性质研究表明菌株S-8所产蛋白酶酸碱稳定性和温度适应性较好,具有进一步开发应用的潜质。 The protease-producing marine bacterial strain S-8 was isolated by transparent circle on the casein-medium from the intestinal tract of healthy octopus and identified by physiological and biochemical experimental,16 S r DNA sequence alignment and scanning electron microscope. The enzyme properties were subsequently investigated. The results showed that:the strain S-8 was identified as vibrio splendid,named vibrio splendid S-8. The bacteria vibrio splendid S-8 showed high protease activity,the activity of crude protease reached 89.12U/m L. The molecular weights of crude protease were 149.6k D,127.4KD,78.9KD,65.1KD,57.3KD and 40.3KD respectively,determined by SDS-PAGE. According to the enzyme property study,strain S-8 produced protease showed a good p H stability and thermal adaptability and thus has good potential for further research and application.
