微小RNA-126在结肠癌组织的表达及临床意义

摘要 目的 探讨微小RNA(miRNA,miR)-126在结肠癌组织中的表达及其临床意义。 方法 采用反转录-聚合酶链反应(RT-PCR)检测miR-126在结肠癌组织的表达并分析其与临床病理特征关系。通过RT-PCR检测人结肠癌SW480细胞及正常结肠上皮细胞FHC中miR-126的表达水平;采用人工合成的miR-126模拟物瞬时转染人结肠癌SW480细胞,采用细胞计数试剂盒(CCK-8)分析转染后细胞增殖变化。 结果 RT-PCR结果显示,结肠癌组织miR-126阳性表达率为38.33%,显著低于癌旁组织(P=0.017);miR-126的低表达与患者的淋巴结转移(P=0.005)、组织分化程度(P=0.018)、肿瘤TNM分期(P=0.000)明显相关,与患者的年龄、性别无明显相关(P=0.350、0.819)。人结肠癌SW480细胞中miR-126的相对表达量为0.355±0.060,较FHC细胞显著降低,差异有统计学意义(P=0.024);采用人工合成的miR-126模拟物转染人结肠癌SW480细胞,SW480细胞中过表达miR-126可显著抑制细胞增殖(P=0.012、0.003、0.000)。 结论 miR-126在结肠癌组织表达下调,影响结肠癌细胞的增殖,抑制结肠癌的发生发展。 Objective To discuss the expression of microRNA (miRNA, miR)-126 in colon cancer and its clinical significance.Methods The expression of miR-126 in colon cancer and cell line was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and its relationship with clinicopathological features was analyzed. We transfected the miR-126 mimics into SW480 cells, and cell counting kit-8 (CCK-8) was used to analyze cell proliferation. Results The positive expression rate of miR-126 in colon cancer was 38.33%, which was significantly lower than that in adjacent normal tissues (P=0.017). The low expression of miR-126 in colon cancer tissue was related to lymph node metastasis (P=0.005), the degree of histological differentiation (P=0.018) and TNM staging (P=0.000), but not to the age and sex of the patients (P=0.350, P=0.819). The relative expression of miR-126 in colon cancer SW480 cells was 0.355±0.060, which was significantly lower than that of FHC cells (P=0.024). Overexpression of miR-126 in SW480 cells could significantly suppress cell proliferation (P=0.012, 0.003, 0.000).Conclusion The low expression of miR-126 in colon cancer can affect the proliferation of colon cancer cells and inhibit the development of colon cancer.
