摘要
目的探讨SDF-1/CXCR4生物轴对口腔鳞状细胞癌癌细胞增殖与侵袭的影响。方法以口腔鳞癌细胞株Tca-8113为研究对象,实验分对照组、SDF-1组和抗体封闭组。通过MTT法检测细胞增殖能力的变化,采用Transwell Chamber检测SDF-1对Tca-8113细胞侵袭及CXCR4封闭对其侵袭的影响。结果基质细胞衍生因子-1(SDF-1)可以明显促进口腔鳞状细胞癌细胞增殖(P<0.05),而抗CXCR4单克隆抗体可明显抑制肿瘤细胞的增殖(P<0.05)。侵袭实验显示,SDF-1可促进Tca-8113细胞的侵袭,50μg/m L抗CXCR4单克隆抗体对CXCR4的封闭能抑制Tca8113细胞的这种侵袭活性,与对照组相比差异有统计学意义(P<0.05)。结论趋化因子SDF-1及其受体CXCR 4相互作用在口腔鳞癌细胞增殖和侵袭中发挥着重要作用,干扰CXCR4/SDF-14的相互作用有可能成为治疗口腔鳞状细胞癌的一个有意义的靶点和策略。
Objective To study the effect of chemokine SDF - 1/CXCRg biologial axis on proliferation and invasion of oral squa- mous carcinoma cells. Method The proliferation of Tca -8113 cells were evaluated by MTT assay in different groups (control group,SDF - 1 group and anti - CXCR4mAb - blocking group. TransweU Chamber was used to detected the invation of Tca - 8113 cells in presence of SDF - 1 or when CXCR4 was blocked by anti - CXCR4mAb in the groups. Results In MTY assay,recombinant SDF - 1 stimulated the proliferation of tumor cells(P 〈9. 05) ,and CXCR4 - monoclonal antibodies decreased the proliferation ( P 〈 O. 05). The chemotactic invasion of Tea - 8113 cells could be induced by SDF - 1 ( P 〈 0. 05) and this induction was significantly reduced by anti - CXCR4mAb (50μg/mL, P 〈 0. 05 ). Conclusion SDF - 1 andCXCR4 may play an important role in the proliferation and invasion of oral squamous cell carcinoma and intervention with the interaction of SDF - 1/CXCR4 could be a meaningful therapeutic target and strategy.
