摘要
目的:探讨转录因子T-bet、GATA-3、Foxp3及STAT-6在支气管哮喘患者中的作用,研究三伏天灸治疗支气管哮喘的免疫调节机理。方法:将18例支气管哮喘患者纳入治疗组,选取18例健康志愿者纳入对照组,运用实时荧光定量PCR检测支气管哮喘患者治疗前、治疗6个月后和对照组样本外周血单个核细胞(PBMNC)中T-bet、GATA-3、Foxp3和相关信号通路STAT-6的mRNA表达,采用流式细胞术检测外周血Th1、Th2比例,酶联免疫吸附法(ELISA)检测IFN-γ、IL-4水平。结果:经三伏天灸治疗后,支气管哮喘患者IFN-γ分泌降低,T-bet、GATA-3表达水平升高,STAT-6水平降低;治疗前后Foxp3水平差异无统计学意义(P>0.05)。结论:三伏天灸可下调STAT-6,上调T-bet、GATA-3,从而抑制Th2过度分泌,调节Th1/Th2比例平衡,在哮喘病程中起到免疫调节作用。
Objective: To investigate the role of transcription factors of T-bet, GATA-3, Foxp3 and STAT-6 in bronchial asthma patients, and study the immunoregulation mechanism of medicinal vesiculation in treatment of bronchial asthma. Methods: 18 cases of bronchial asthma patients were included in treatment group and 18 cases of healthy volunteers in control group; for the PBNMC samples of bronchial asthma patients those before and 6 months after treatment and control group, real-time PCR was used to detect the mRNA expression of T-bet, GATA-3, Foxp3 and related signaling pathway STAT-6, flow cytometry to detect Th1/Th2 ratio and ELISA to detect IFN-γ, and IL-4 levels. Results: After medicinal vesiculation treatment, IFN-γ secretion in bronchial asthma patients had decreased, expression levels of T-bet and GATA-3 had increased and STAT-6 level decreased; there was no significant difference between Foxp3 level that before and after treatment (P〉0.05). Conclusion: Medicinal vesiculation can cut down STAT-6 and raise T-bet and GATA-3, thus inhibiting excessive secretion of Th2 and regulating the balance of Th1/Th2 ratio, which plays an immunoregulation role in course of asthma.
