摘要
目的:探讨人肠癌组织中Survivin与p-m TOR蛋白表达情况,并分析两者相关性。方法:首先应用免疫组织化学法检测86例肠癌组织中Survivin与p-m TOR蛋白的表达;然后利用Western blot技术检测4对冰冻肠癌组织及其癌旁组织中Survivin与p-m TOR蛋白的表达情况。结果:在免疫组化实验中,肠癌组织中Survivin蛋白的表达与组织学分级、淋巴结转移以及Dukes分期呈正相关(P<0.05),与年龄、性别以及肿瘤大小无关(P>0.05);而p-m TOR蛋白的表达与淋巴结转移和组织学分级也呈正相关(P<0.01),与年龄、性别、肿瘤大小以及Dukes分期无关(P>0.05)。不仅如此,Survivin与p-m TOR在组织中的表达水平呈密切正相关。在Western blot实验中,肠癌组织Survivin与p-m TOR蛋白表达水平均明显高于癌旁组织,进一步说明Survivin与p-m TOR在肠癌组织中的表达水平上升且呈正相关。结论:肠癌组织中Survivin与pm TOR蛋白的阳性表达与多项临床病理参数相关,并且两者表达也密切相关,提示其在肠癌进展中起到了重要作用,为探索肠癌诊疗新靶点提供理论依据。
Objective:To investigate the expression of Survivin and p - roTOR proteins in colorectal cancer and analyze the correlation of these two proteins. Methods: Immunohistochemical technique was used to examine the ex- pression of Survivin and p - mTOR proteins in 86 cases of colorectal cancer tissues. Relationship between the expres- sion of Survivin and p - mTOR proteins and clinicopathological parameters were analyzed. Results: Elevated Survivin expression in colorectal cancer tissues was correlated with histological grade, lymph node metastasis and Dukes stage (P 〈 0.05 ). High p - roTOR expression in colorectal cancer tissues was also correlated with lymph node metastasis and histological grade (P 〈 0.01 ). Besides, high Survivin tended to indicate high p -roTOR expression from our cor- relation analysis. Conclusion: Survivin and p - roTOR were related to several important clinicopathological factors. Thus, they may serve as promising targets for therapy of colorectal cancer.
