紫草素对人肝癌HepG2细胞顺铂增敏作用的研究 被引量:2

摘要 目的研究紫草素对人肝癌HepG2细胞顺铂增敏作用并初探其可能的作用机制。方法取对数生长期人肝癌HepG2细胞,设空白对照组、紫草素(0.5μg/ml)组、顺铂(40μg/ml)组和联合(紫草素0.5μg/ml+顺铂40μg/ml)组,均设10个复孔;检测细胞增殖抑制率,细胞周期和细胞凋亡状况,Bax mRNA、bcl-2 mRNA表达,caspase-3蛋白表达。结果与顺铂组比较,联合干预组HepG2细胞增殖抑制率显著升高,细胞周期G_0/G_1期显著延长,细胞凋亡率显著升高,Bax/bcl-2比值显著升高,caspase-3蛋白表达上调。结论紫草素具有提高人肝癌HepG2细胞顺铂敏感性的作用,其机制可能与紫草素阻滞细胞周期及调节凋亡相关基因和蛋白表达有关。 Objective To investigate the efficacy enhancing effect of Shikonin on human liver cancer HepG2 cells treated by Cisplatin. Methods The human liver cancer HepG2 cells in logarithmic growth phase was treated with Shikonin (0.5 μg/ml), Cisplatin (40 μg/ml) and Shikonin (0.5 μg/ml) + Cisplatin (40 μg/ml), and set blank group, n = 10. The cellular growth inhibition rate, cell cycle and apoptosis rate, the expression of Bax mRNA, bcl-2 mRNA and caspase-3 protein were detected. Results Compared with Cisplatin group, the cellular growth inhibition rate in Shikonin + Cisplatin group was significantly increased, the cell cycle was arrested at Go/ G1 phase, the AI was significantly increased, the ratio of Bax/bcl-2 was significantly increased, the expression of caspase-3 protein was significantly increased. Conclusions Shikonin can effectively enhance the effect on human liver cancer HepG2 cells treated by Cisplatin, which perhaps related to its effects of altering the cell cycle and the expression of apoptosis-related genes and proteins.
