基于SRAP分子标记的广宁红花油茶种质资源遗传多样性研究 被引量:17

摘要 利用SRAP分子标记建立广东省4个种群153个单株的广宁红花油茶种质资源遗传多样性评价体系,从594对引物组合中最终选出13对引物,结果表明:13对引物组合共扩增出206条带,平均每对引物组合获得15.85条,多态性条带为187条,多态性条带比率(PPB)为90.78%。种群Nei's基因多样度(h)、Shannon多样性指数(I)、多态性位点百分比(PPL)变化范围分别为0.217 9~0.356 3,0.321 7~0.524 8及58.25%~92.72%,平均值分别为0.307 8,0.456 3及83.62%,表明广宁红花油茶具有丰富的遗传多样性。聚类分析表明,当阈值为0.93时,可将广宁红花油茶4个种群分为2大类,广东清远单独成为一类,第二类包括广东广宁县,广东增城,广东云浮。广东云浮与广东增城遗传距离为0.019 3,遗传距离最小,说明二者的亲缘关系最近。AMOVA分子变异方差分析显示,在广宁红花油茶总的遗传变异中,种群内占95%,而种群间仅占5%。 In order to establish the evaluation system of genetic diversity of 153 individule tree from 4 populations in Camellia semiserrata Chi of Guangdong Province, 13 pairs of SRAP primers were selected from 594 combinations and applied to analyze the genetic diversity. The results showed that a total of 206 fragments was amplified, and the average amplified fragments of each primer pair were 15.85. There were 206 polymorphic bands, and the percentage of polymorphic bands(PPB) was 90.78%. The Nei's gene diversity(h), Shannon's information index(I) and percentage of polymorphic loci(PPL) of different populations were 0.2179-0.3563, 0.321 7-0.524 8 and 58.25 %-92.72 %, with the average 0.307 8, 0.456 3 and 83.62 %, respectively, showing rich genetic diversity at population of C. Semiserrata. The dendrogram indicated that when the threshold was 0.93, 4 populations of C. semiserrata was divided into two major categories that Qingyuan population was one group and the second group included populations of Guangning County, Zengcheng and Yunfu. Genetic distance of Yunfu and Zengcheng was minimum(0.0193) showing a closer relationship for these two populations. The analysis of molecular variance(AMOVA) showed the 95% of the genetic variation existed within populations and 5% among them.
