替考拉宁产生菌A. teichomycoticus FIM-37发酵条件优化 被引量:4

Optimization of Fermentation Conditions of Teicoplanin-Producing Strain A. teichomyceticus FIM-37

摘要 为了优化替考游动放线菌FIM-37菌株产替考拉宁发酵条件,采用单因素试验和正交试验研究发酵培养基组成,探讨摇瓶发酵的主要影响因子初始pH值、装液量、转速、温度等发酵参数的影响。结果确定了适宜发酵培养基和发酵参数:可溶性淀粉3.0%,葡萄糖1.0%,黄豆粉4.5%,蛋白胨0.5%,磷酸氢二钾0.02%,氯化镁0.05%,硫酸铵0.02%,脯氨酸0.1%,碳酸钙0.3%,初始pH 7.5~8.0,装液量50 mL/500 mL,种龄40~48 h,接种量5%,摇床转速230 r/min,发酵温度30℃,发酵时间8 d。优化后的发酵水平较原工艺提高了40%以上,该优化发酵工艺为替考拉宁的工业化生产奠定了基础。 To optimize the fermentation conditions of teicoplanin producing strain A. teichomyceticus FIM-37,the single factor experiments and orthogonal experiments were conducted to study the liquid fermentation medium and parameters of A. teichomyceticus FIM-37 fermentation. It was determined that the appropriate fermentation medium formula composition and optimal fermentation parameters were as follows: soluble starch 3. 0%,glucose 1. 0%,soybean flour 4. 5%,peptone 0. 5%,K_2HPO_4 0. 02%,and MgCl_2 0. 05%,( NH_4)_2SO_4 0. 02%,proline 0. 1%,CaCO_3 0. 3%,initial pH at 7. 5 ~ 8. 0,filling volume 50 mL for a 500 mL shake flask,cultivation time of seed 40 ~ 48 h,inoculum size 5%,shaker's rotating speed 230 r/min,fermentation temperature 30 ℃,and fermentation time 8 d. The teicoplanin yield of A. teichomyceticus FIM-37 on the optimal fermentation conditions was enhanced more than 40% higher than that of the original. It laid a foundation for producing teicoplanin on industrial scale.
