摘要
目的:建立防风配方颗粒的HPLC含量测定方法,为防风配方颗粒的质量评价提供依据。方法:采用高效液相色谱法测定配方颗粒中升麻苷、升麻素、5-O-甲基维斯阿米醇苷、亥茅酚苷的含量,色谱柱为X-Bridge C18,流动相为乙腈-0.3%磷酸水溶液梯度洗脱,流速为1.0 mL/min,检测波长为220 nm,进样量为10μL。结果:升麻素、5-O-甲基维斯阿米醇苷、亥茅酚苷,升麻苷检测进样量线性范围为分别为0.21~4.12μg/mL(R=0.998 5),0.83~16.52μg/mL(R=0.998 3),0.31~6.20μg/mL(R=0.998 7),0.24~4.76μg/mL(R=0.998 2);精密度、稳定性、重复性试验的RSD<5%;加样回收率分别为97.82%(RSD=2.44%),95.18%(RSD=1.88%),99.33%(RSD=3.70%),95.45%(RSD=4.30%)。结论:HPLC法操作简便、稳定、重复性好,可用于防风配方颗粒中有效成分含量的测定。
Objective :To establish the method for determinating 4 ingredients in formula granules of Saposhnik- oviae radix by HPLC, in order to provide the basis for quality evaluation of formula granules of Saposhnikoviae ra- dix. Methods : The contents of prim - O - glucosylcimifugin, cimifugin, 4"- 0 - 15 - D - glucosyl - 5 - O - methyl- visarnmioside and see - o - glucosylhamaudol in formula granules were determinated by HPLC. The chromatograph- ic column was X -Bridge C18; the mobile phase was aeetonitrile -0.3% phosphoric acid solution for gradient elution;the flow rate was 1 mL/min; the detection wavelength was 220 nm; and injection volume was 10 μL. Results:The linear ranges were respectively 0.21 -4.12 μg/mL( R = 0.998 5 ) for cimicifugin,0.83 - 16.52 μg/ mL( R = 0.998 3 ) for prim - O - glucosylcimifugin, 0.31 - 6.20 μg/mL( R = 0.998 7 ) for 4'- O - 15 - D - glu- cosyl - 5 - O - methylvisamminol, and 0.24 - 4.76 μg/mL ( R = 0.998 2) for sec - O - glucosylhamaudol. RSDs of precision, stability and reproducibility tests were lower than 5 %. The recovery rates of injection were respectively 97.82% (RSD = 2.44%, n = 5 ) for cimicifugin, 95.18 % (RSD = 1.88 %, n = 5 ) for prim - O - glucosyl- cimifugin, 99.33 % ( RSD = 3.70% , n = 5 ) for 4"- O - 15 - D - glucosyl - 5 - 0 - methylvisamminol, and 95.45 % ( RSD = 4.30%, n = 5 ) for see - O - glucosylhamaudol. Conclusion : This method is simple, stable and better reproducible, and can be used for simultaneous determination of contents of active ingredients in Saposhnikoviae ra- dix and its formula granules.
