过渡金属铜修饰的仲钨酸盐抑制人卵巢癌细胞增殖的研究 被引量:2

Inhibitory effect of a paradodecatungstate-B compound decorated by transition metal copper on the growth of human ovarian carcinoma SKOV-3 cells

摘要 研究了过渡金属铜修饰的仲钨酸盐化合物[Na_2(H_2O)_(10)][Cu_4(H_2O)_(12)(H_2W_(12)O_(42))]·15H_2O(CuW_(12))的体外抗肿瘤活性.取人卵巢癌细胞SKOV-3加入不同浓度CuW_(12)处理,应用四甲基偶氮唑盐(MTT)比色法分析细胞增殖,计算了半数抑制浓度(IC_(50));采用光学显微镜观察SKOV-3细胞的凋亡形态变化;分析了细胞周期和细胞凋亡,计算了各期细胞比例及细胞凋亡率.结果表明:CuW_(12)对SKOV-3细胞增殖抑制的IC_(50)值为45.2μmol/L,且呈剂量依赖性;与经典的仲钨酸盐Na_(10)(H_2W_(12)O_(42))·26H_2O(NaW_(12))相比,过渡金属铜修饰的仲钨酸盐抗肿瘤活性更强;光学显微镜下观察到CuW_(12)处理组细胞数目明显减少,出现变圆、缩小、老化等形态变化;不同浓度CuW_(12)处理12h后早期凋亡细胞所占的百分比显著增加且呈剂量依赖性. This paper has studied the in vitro anti-tumor activity of a novel paradodecatungstate-B compound decorated by transition metal copper:[Na2(H_2O)_(10)][Cu_4(H_2O)_(12)(H2W_(12)O_(42))]·15H_2O(abbreviated form:CuW_(12)).Human ovarian carcinoma SKOV-3 cell was treated with different concentration of CuW_(12),then its IC_(50) was measured by MTT assay.The morphological change in SKOV-3 cell was observed by optical microscopes.Flow cytometry was applied to analyze cell cycle and apoptosis by computing the ratio of SKOV-3 cells in different phase and its apoptotic ratio.The results show that CuW_(12) can inhibit SKOV-3cell proliferation with IC_(50) of 45.2μ mol/L,showing dosedependent.Compared with CuW_(12) and classic paradodecatungstate compound Na_(10)(H2W_(12)O_(42))·26H_2O(abbreviated form:NaW_(12)),CuW_(12) exhibits higher inhibitory effect owing to the introduction of copper atom.Under the optical microscopes,we observed the number of SKOV-3 cells treated with CuW_(12) decreased significantly,cell rounding,shrinking and aging.Treated with different concentrations of CuW_(12) after 12 h,the proportions of SKOV-3 cells on the early apoptosis increased markedly,showing dose-dependent.
