摘要
目的:建立同时测定五味沙棘颗粒中甘草苷和甘草酸含量的方法。方法:采用高效液相色谱法。色谱柱为Thermo scientific ODS-2 Hypersil,流动相为乙腈-0.5%磷酸溶液(梯度洗脱),流速为1.0 m L/min,检测波长为237 nm,柱温为25℃,进样量为20μL。结果:甘草苷和甘草酸检测进样量线性范围分别为48~480μg(r=0.999 9)、90~900μg(r=0.999 9);定量限分别为0.55、2.10μg/m L,检测限分别为0.16、0.62μg/m L;精密度、稳定性、重复性试验的RSD<2.0%;加样回收率分别为95.08%~97.58%(RSD=0.93%,n=6)、95.86%~99.89%(RSD=1.67%,n=6)。结论:该方法简单准确、重复性好,适用于五味沙棘颗粒中甘草苷和甘草酸含量的同时测定。
OBJECTIVE: To establish a method for simultaneous determination of liquiritin and glycyrrhizic acid in Wuwei sha- ji granules. METHODS: HPLC method was adopted. The determination was performed on Thermo scientific ODS-2 Hypersil col- umn with mobile phase consisted of acetonitrile-0.5% phosphoric acid (gradient elution) with flow rate of 1.0 mL/min. The detec- tion wavelength was set at 237 nm, and column temperature was 25 ℃. The sample size was 20μL. RESULTS: The linear ranges of liquiritin and glycyrrhizic acid were 48-480 μg(r=0.999 9) and 90-900 μg(r=0.999 9), respectively. The limits of quantitation were 0.55, 2.10 μg/mL, and the limits of detection were 0.16, 0.62 μg/mL. RSDs of precision, stability and reproducibility tests were all lower than 2.0%. The recoveries were 95.08%-97.58% (RSD=0.93%, n=6)and 95.86%-99.89% (RSD= 1.67% ,n=6), respectively. CONCLUSIONS: The method is simple, accurate and reproducible, and can be used for the content determination of liauiritin and glvcvrrhizic acid in Wuwei shaii granules.
