摘要
【目的】探明PRNP基因表达量与鸡马立克氏病发病之间的关系.【方法】从甘肃省康乐县某养鸡场采集马立克(MD)病鸡组织样品,通过RT-PCR检测MDV致瘤相关基因Meq和pp38以及病理组织学观察将样品分为MD肿瘤病变鸡、MD无病变鸡与健康鸡,最后通过荧光定量PCR方法分析3组中PRNP基因mRNA水平的相对表达量的差异.【结果】MD无病变鸡中PRNP基因的表达规律与健康鸡基本一致,但与MD肿瘤病变鸡完全不同;除脑外,MD病鸡组织PRNP基因的表达量均高于健康鸡,其中肿瘤病变组织PRNP基因的表达量显著高于无病变组织(P<0.05),而且PRNP基因在MD病鸡肿瘤病变组织与健康鸡组织的表达差异与肿瘤病变程度有关.【结论】PRNP基因的表达量与MDV感染存在相关性,PRNP基因在MD肿瘤组织中过表达,且PRNP基因的差异表达与肿瘤病变程度有关.
【Objective】In order to research the relationship between the expression of PRNPgene and the pathogenesis of Marek's disease in chickens.【Method】The diseased tissues of chickens infected with MD were collected from a chicken farm in Kangle county of Gansu province.Samples were divided into neoplastic lesions,no neoplastic lesions and health chickens through the methods of RT-PCR to detect the tumor-related genes Meq and pp38 and histopathological observations.The relative expression of PRNP gene mRNA levels were analyzed in the three groups by Real-time PCR.【Result】The expression rule of PRNPgene in non-pathological chickens showed the same as healthy chickens,but was different from pathological chickens.Except brain,PRNPgene expression quantity in affected chickens was higher than that in healthy chickens.PRNP gene expression quantity in MD tumor tissues was significantly higher than non-pathological tissues(P〈0.05).Moreover,the expression differences of PRNP gene between tumor tissues infected with MD and normal tissues was related to the lesion degree.【Conclusion】PRNP gene expression is related to the infection of MDV,PRNPoverexpression in tumor tissues,and the differential expression of PRNPgene in various neoplastic tissues is related to the lesion degree.
