摘要
通过分析金针菇(Flammulina velutipes)子实体原基期、伸长期、成熟期的转录组数据,得到一个差异表达基因fv-msf1。对其结构、序列特征及表达情况进行分析,结果显示该基因全长2709 bp,包含13个外显子,12个内含子,开放阅读框长1770 bp,编码589个氨基酸。结构域分析表明该基因编码的蛋白含主要协助转运蛋白超家族(major facilitator superfamily,MFS)保守结构域。通过实时荧光定量PCR(real-time quantitative fluorescence PCR,q-PCR)对子实体不同发育阶段以及菌盖不同发育时期的表达量进行分析,结果显示该基因在金针菇伸长期的菌盖(尤其是0.3~0.9cm)中高表达,由此推断该基因在金针菇菌盖发育中发挥一定作用。
Through comparative transcriptome analysis of Flammulina velutipes at different developmental stages of fruit body(primordial stage,elongation stage,and maturation stage),a differentially expressed gene fv-msf1was identified.Sequence analysis indicated that fv-msf1was 2709bp in length,contained 13exons and 12introns,and had an open reading frame of 1770bp encoding 589amino acids.Domain analysis indicated this gene contained a major facilitator superfamily conservative domain.Using real-time quantitative fluorescence PCR,the gene transcription levels of fv-msf1at different developmental stages of fruit body and pileus were measured respectively.The results showed that fv-msf1was intensively transcribed in pileus at elongation stage(pileus diameter 0.3~0.9cm),suggesting its role in pileus development of F.velutipes.
