应用毛细管区带电泳-紫外检测法分离猪肉中的4种核苷酸 被引量:1

Determination of four nucleotides in pork by capillary zone electrophoresis with direct ultraviolet detection

摘要 应用毛细管区带电泳-紫外检测器(CZE-UV)法同时分离测定猪肉中4种核苷酸。采用阴极检测方法,考察了硼砂浓度、运行缓冲液浓度及p H值、分离电压、毛细管温度4个因素对分离效果和重现性的影响。试验得出最优分离条件为:分离电压25 k V、检测波长214 nm、毛细管温度25℃,以12 mmol/L的硼砂(p H10.0)为运行缓冲液,4种核苷酸在13 min内达到基线分离;检出限最低可达0.4 mg/L;线性范围为(20~100)mg/L;相关系数(R^2)均大于0.997;日内精密度为0.37%~3.97%;日间精密度为1.29%~10.8%。实际猪肉样品中4种核苷酸的加标回收率为103.69%~107.09%。结果表明CZE-UV方法简单、快速、实用,可为猪肉质量评估提供借鉴方法。 A method of capillary zone electrophoresis(CZE) with direct ultraviolet(UV) detection was developed for the separation and determination of four nucleotides in pork. The effects of sodium tetraborate concentration, p H of running buffer, separation voltage and capillary temperature on the reproducibility and ef ficiency were investigated by using negeative detection. The optimum electrophoretic conditions were as follows: 12 mmol/L sodium tetraborate(p H10.0) as running buffer, Capillary temperature was 25 ℃, applied voltage of 25 k V, detection wavelength of 214 nm. The base line separation of the four nucleotides was achieved successfully within 13 min. The lowest detection limit was 0.4 mg/L. The analyses showed the four nucleotides linearities within(20~100)mg/L, correlation coefficients(R^2) was not less than 0.997. The Intra-day relative standard deviations of migration time and peak area were 0.37%~3.97%, respectively. The Inter-day relative standard deviations of migration time and peak area were respectively 1.29%~10.8%. The recoveries of the four nucleotides spiked in a real sample were between 103.69%~107.09%. In conclusion, the CZE-UV method could be used to provide reference for pork quality evaluation and this method is simple, rapid and practical.
