RASSF1A基因启动子区甲基化状态在食管鳞癌诊断中的研究 被引量:5

摘要 目的:检测RASSF1A(ras association domain family 1A,RASSF1A)基因在肿瘤组织的甲基化频率差异,并且评价其对食管鳞癌的诊断价值。方法:收集食管鳞癌组织、癌旁组织各43例,记录病人的病理资料。应用甲基化特异性PCR法结合琼脂糖凝胶电泳检测RASSF1A基因在组织中的甲基化情况,比较甲基化基因检测与肿瘤标志物两种检测标志物在食管癌诊断中的价值。将实验结果录入统计软件,计数资料两组间比较采用χ~2检验统计分析。结果:RASSF1A基因在肿瘤组织中的甲基化频率为34.88%,显著高于癌旁组织6.98%,差异有统计学意义(P=0.001);RASSF1A基因启动子区异常甲基化状态与患者年龄、性别、肿瘤大小、分化程度、肿瘤深度、是否淋巴结转移及临床分期无明显关系(P>0.05),RASSF1A基因甲基化检测阳性检测率为34.88%,明显高于肿瘤标志物的阳性检测率11.62%,差异有统计学意义(P=0.011);结论:RASSF1A基因启动子区甲基化的检测在食管鳞癌的诊断中具有较高的价值。 Abstract Objective: To investigate the difference in methylation frequency of Ras association domain family 1A (RASSF1A) gene in tumor tissue and its value in the diagnosis of esophageal squamous cell carcinoma. Methods: A total of 43 esophageal squamous cell carcinoma tissue samples and 43 adjacent tissue samples were collected, and the patients' pathological data were recorded. Methylation-specific PCR and agarose gel electrophoresis were used to measure the methylation of RASSF1A gene, and the value of methylated gene detection versus tumor markers in the diagnosis of esophageal cancer was compared. The experimental results were entered into the statistical software, and the chi-square test was used for the comparison of categorical data between groups. Results: The tumor tissue samples had a significantly higher methylation frequency of RASSF1A gene than the adjacent tissue samples (34.88% vs 6.98%, P = 0.001). Abnormal methylation of RASSF1A gene promoter region was not significantly associated with age, sex, tumor size, degree of tumor differentiation, tumor depth, presence or absence of lymph node metastasis, and clinical stage (P 〉 0.05). Methylated gene detection achieved a significantly higher positive rate of RASSF1A gene methylation than detection of tumor markers (34.88% vs 11.62%, P = 0.011). Conclusions: Detection of methylation of RASSF1A gene promoter region has a high value in the diagnosis of esophageal squamous cell carcinoma.
