阴地蕨药材质量标准研究

摘要 目的对阴地蕨药材进行生药鉴定研究,以制定其药材的质量标准。方法观察阴地蕨药材的性状和显微鉴别特征;以木犀草素为对照品进行TLC色谱研究;采用紫外分光光度法测定总黄酮,高效液相色谱法测定木犀草素含量;并检查其水分、总灰分、酸不溶性灰分、醇溶性浸出物。结果 TLC采用硅胶G板,以环己烷-乙酸乙酯-甲醇-甲酸(20︰10︰3︰1.5)为展开剂,1%FeCl_3乙醇溶液为显色剂,可见清晰的斑点;紫外分光光度法以芦丁为对照品,以总黄酮含量为指标,对8批药材测定,药材中阴地蕨总黄酮含量不低于12.55 mg·g^(-1);HPLC法以木犀草素为对照品,以甲醇-水(含0.5%冰醋酸)为流动相,检测波长350 nm,对8批药材测定,药材中阴地蕨木犀草素含量不低于13.75μg·g^(-1)。结论阴地蕨药材的显微、TLC鉴别、总黄酮及木犀草素的含量测定方法准确可靠,可有效控制阴地蕨药材的质量。 Objective To determine the quality standard of Botrychium ternatum(Thunb.) Sw.. Methods The macroscopic and microscopic characteristics were observed. Luteolin was used as the reference substance in the TLC identification. UV was used to determine the content of total flavonoids. HPLC was used to determine the content of luteolin. The water content, total ash, acid-insoluble ash, and ethanol soluble extraction were checked. Results The TLC method used G taking cyclohexane-aceticether-methyl alcohol-formic acid(20 ︰ 10 ︰ 3 ︰1.5) as the developer and ethanol solution of 1% ferric chloride as the chromogenic reagent, and clear spots were shown. The UV method took rutinum as the reference substance, and the content of total flavonoids as the index. The content of samples of 8 batches were determined, and the lowest content of total flavonoids was 12.55 mg·g^(-1). The HPLC method took luteolin as the reference substance, and methyl alcohol-water(containing 0.5% acetic acid) as the mobile phase. The detection wavelength was 350 nm. The content of samples of 8 batches were determined, and the lowest content of luteolin was 13.75 μg·g^(-1). Conclusion This method is reliable and accurate, and can be used for the quality control of Botrychium ternatum(Thunb.) Sw..
