强力霉素对骨关节炎大鼠膝关节软骨成纤维细胞生长因子-18表达的影响

摘要 目的探讨强力霉素对骨关节炎大鼠膝关节软骨成纤维细胞生长因子-18(FGF-18)表达的影响。方法32只SD大鼠通过膝关节前交叉韧带离断建立骨关节炎模型,随机分为A、B、C、D组,分别每日1次腹膜腔注射0.9%氯化钠溶液1 mL及3、6、9 mg/(kg·d)强力霉素溶液,共8周;免疫组化染色检测股骨髁FGF-18表达。结果强力霉素处理组FGF-18表达低于对照组(P<0.05),而不同剂量强力霉素组间比较差异无统计学意义(P>0.05)。结论强力霉素可减少骨关节炎大鼠膝关节软骨FGF-18表达。 Objective To investigate the effect of doxycycline on fibroblast growth factor-18 (FGF-18) expression of knee articular cartilage in rats with osteoarthritis. Methods The knee osteoarthritis models of 32 SD rats were established using detachment of the anterior cruciate ligament, and then randomized to A- D groups. Four groups respectively received intraperitoneal injection with 0.9% sodium chloride (lmL) and doxycycline solution (3, 6, and 9 mg/(kg.d) once daily for 8 weeks. FGF-18 expression of femoral condyle was determined by immunohistochemistry. Results FGF-18 expression was lower in doxycycline-treated groups than in control group (P〈0.05), but its expression showed no difference among doxycycline-treated groups (P〉0.05). Conclusion Doxycycline can reduce FGF-18 expression of the knee articular cartilage in rats with osteoarthritis.
