摘要
目的研究STAT3基因以及其通路对多形性成胶质细胞瘤的作用。方法利用免疫组织化学检测胶质瘤组织中STAT3的激活情况;利用蛋白印迹免疫杂交检测胶质瘤细胞系中STAT3的磷酸化;加入药物葫芦素与WP1066,观察药物对STAT3的影响,分析对胶质瘤存活的影响;用shRNA敲除STAT3基因,分析对胶质瘤细胞生长的影响。结果胶质瘤组织和胶质瘤细胞系中STAT3通路被磷酸化激活;STAT3信号受到葫芦素与WP1066的抑制,可以降低胶质瘤的体外存活与增殖,葫芦素与WP1066促进胶质瘤细胞的凋亡;shRNA敲除STAT3基因使得胶质瘤细胞在体外生长减慢。结论证明STAT3信号对于神经胶质瘤的存活、增殖与肿瘤发生都有非常重要的影响,抑制STAT3信号通路会导致肿瘤细胞的凋亡。
Objective To study the role of STAT3 gene and its pathway in glioblastoma multiforme.Methods Immunohistochemistry was used to investigate the activation of STAT3 in glioma tissue while Western blot hybridization assay was used to investigate the phosphorylation of STAT3 in glioma cell lines. Cucurbitacin and WP1066 were added to observe the effects of drugs on STAT3 and the survival of glioma. ShRNA was used to knockdown STAT3 gene to analyze the impact of STAT3 on glioma cells.Results In glioma tissue and glioma cell lines,STAT3 pathway was activated while STAT3 signals were inhibited by cucurbitacin and WP1066,which caused the decline of survival and proliferation for glioma in vitro. Moreover,cucurbitacin and WP1066 promoted apoptosis of glioma cells. Knockdown of STAT3 slowed down glioma growth in vitro.Conclusion STAT3 signaling has a very important impact on the survival,proliferation and tumorigenesis of glioma. Inhibiting STAT3 signaling pathway can lead to apoptosis of tumor cells.
