姜黄素对溃疡性结肠炎模型小鼠结肠组织TNF-α、IL-1β、IL-10表达的影响 被引量:8

摘要 目的观察姜黄素对溃疡性结肠炎(UC)模型小鼠结肠组织肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素10(IL-10)表达的影响,探讨其治疗UC的可能作用机制。方法SPF级健康雄性BALB/c小鼠60只,随机分为正常对照组、模型组、姜黄素组、柳氮磺胺吡啶(SASP)组,各15只。采用右旋葡聚糖硫酸钠(DSS)法复制UC小鼠模型,腹腔注射相应药物,连续7天。观察小鼠一般生理状态,疾病活动度指数(DAI)评分。末次给药后处死小鼠,行结肠大体形态损伤指数(CMDI)评分,组织切片染色,光镜下观察结肠组织的病理学变化并评估组织损伤指数(TDI)。ELISA法检测小鼠结肠组织促炎因子TNF-α、IL-1β和抗炎因子IL-10表达水平。结果(1)模型组小鼠大体形态、组织病理均符合溃疡性结肠炎特点。(2)各组DAI评分比较:SASP组[(0.51±0.40)分]、姜黄素组[(0.52±0.36)分]与模型组[(1.38±0.61)分]比较,P<0.01。(3)各组CMDI评分比较:SASP组[(2.14±0.92)分]、姜黄素组[(2.19±1.10)分]与模型组[(3.83±1.44)分]比较,P<0.01。(4)各组TDI评分比较:SASP组[(5.48±1.01)分]、姜黄素组[(6.48±1.03)分]与模型组[(8.89±0.88)分]比较,P<0.01。(5)各组小鼠结肠组织TNF-α、IL-1β及IL-10含量比较:SASP组[(126.75±6.16)pg/m L、(157.51±20.43)pg/m L、(156.33±16.51)pg/m L]、姜黄素组[(254.83±28.29)pg/m L、(236.83±25.21)pg/m L、(136.66±16.49)pg/m L]与模型组[(418.19±41.67)pg/m L、(401.11±35.32)pg/m L、(33.96±6.68)pg/m L]比较,P<0.01。结论姜黄素可以显著改善UC小鼠症状、病理形态,上调小鼠结肠组织IL-10表达水平,抑制TNF-α、IL-1β表达水平,达到对UC的抗炎作用。 Objective To investigate the effects of curcumin on the expression of tumor necrosis factor α(TNF-α), interleukin 1β(IL-1β) and interleukin 10(IL-10) in colon tissue ofmice with ulcerative colitis(UC), and to explore the possible underlying mechanism. Methods Sixty SPF grade healthy BALB/c mice were randomly divided into four groups: blankgroup, model group, curcumintreatmentgroup, salazosulfapyridine(SASP) group.UC model was established by dextran sulphate sodium, and then daily intraperitoneal injected with the corresponding drugs for7 days. During the course of experiment, the general conditions of mice were observed, and disease activity index(DAI) were assessed. At the end of treatment, colon tissue was dissected to evaluate the macroscopic damage index(CMDI) and tissue damage index(TDI) by light microscope and to determine the expression of TNF-α, IL-1βand IL-10 by ELISA. Results General morphology and histopathology of the model group conforms to the manifestations of ulcerative colitis. The DAI score in SASP group, curcumin group was significant different compared with that in model group(0.51±0.40, 0.52±0.36 vs 1.38±0.61, P〈0.01). The CMDI score was statistically difference among model group, SASP group,andcurcumin group(3.83±1.44, 2.14±0.92 and 2.19±1.10, P〈0.01). The TDI score was decreased in SASP group and curcumin group compared to model group(5.48±1.01, 6.48±1.03 vs 8.89±0.88,P〈0.01). The contents of TNF-α, IL-1βand IL-10 in colon tissue in model group(418.19±41.67pg/m L, 401.11±35.32pg/m L, 33.96±6.68pg/m L) also had significant difference compared with those in SASP group(126.75±6.16pg/m L, 157.51 ±20.43pg/m L, 156.33 ±16.51pg/m L) and curcumin group(254.83 ±28.29pg/m L, 236.83 ±25.21pg/m L,136.66±16.49pg/m L), with all P 0.01. Conclusion Curcumin can significantly improve the symptoms and pathological morphology of UC mice by regulating the expression of IL-10 and inhibiting the expression of TNF-α and IL-1β, through which achieving the anti-inflammatory effect of UC.
