自荧光壳聚糖人工抗体的合成及其对Cd(Ⅱ)、Pb(Ⅱ)的痕量检测

摘要 分别以Cd(Ⅱ)、Pb(Ⅱ)为模板分子,壳聚糖为功能单体,戊二醛为交联剂,用油包水(W/O)方法合成了自荧光壳聚糖人工抗体。探讨了pH、交联时间、交联剂用量、振荡器转数、油相和水相体积比对自荧光壳聚糖人工抗体的影响,得到制备自荧光壳聚糖人工抗体的适宜条件:pH≈8、交联时间5 h、戊二醛的量2.5 mL、振荡器转数250 r·min^(-1)、油相和水相的体积比8∶3。同时探讨了金属离子自荧光壳聚糖人工抗体对Cd(Ⅱ)、Pb(Ⅱ)的检测效果。结果表明,自荧光壳聚糖人工抗体实现对Cd(Ⅱ)和Pb(Ⅱ)高选择性识别,高敏感性检测,检出限为10-9mol·L^(-1)。 The self fluorescent chitosan artificial antibody were synthesized by the method of water in oil (W/O) through the copolymerization of using Cd(Ⅱ) or Pb(Ⅱ) as template molecule,chitosan as functional monomer and glutaraldehyde as crosslinking agent,respectively.The effects were investigated to consider the pH value,crosslinking time,the amount of crosslinking agent,the number of the oscillator and the ratio of oil and water phase as impact factors during the synthesis process of the self fluorescent chitosan artificial antibody,and the values of the optimum conditions for its synthesis obtained are 8 (pH),5 h (crosslinking time),2.5 m L (the amount of crosslinking agent),250 r·min^(-1) (the number of the oscillator),8∶3 (the ratio of oil and water phase),respectively.Moreover,the self fluorescent artificial antibody was explored to recognize and detect Cd(Ⅱ) or Pb(Ⅱ) by entering the recognition site and the decrease of fluorescence intensity.Results demonstrate the artificial antibody has the trace detection toward Cd(Ⅱ) and Pb(Ⅱ) with highly selective recognition and high sensibility,respectively,and the detection limit is 10-9mol·L^(-1).
