摘要
为有效保护药用植物桔梗的种质资源,以桔梗种子消毒后萌发的无菌苗为外植体,进行组培快繁技术研究。结果表明:接种诱导培养基以MS+BA1.0 mg·L^(-1)+IBA0.2 mg·L^(-1)最佳,诱导率为83.8%;继代培养基以WPM+ZT1.0 mg·L^(-1)+NAA0.1 mg·L^(-1)最佳,增殖系数为8.82;生根培养基以WPM+IBA0.5 mg·L^(-1)为最佳,生根率达100%,平均生根条数5.21;试管苗炼苗后移栽到水苔:黄心土=1:2的基质上,移栽成活率可达98.21%。
Using germinated sterile plantlets as explants, tissue culture of medicinal plant Platycodon grandiflorum were carried out. The result showed that MS + BA1.0 mg·L^-1 + IBA0.2 mg·L^-1 was optimum for shoot induction with shoot induction rate 83.8%, and WPM + ZT1.0 mg·L^-1 + NAA0.1 mg·L^-1 with proliferation coefficient 8.82 was optimum for subculture. When shoot were cultured on WPM + IB A0.5 mg·L^-1,root induction rate could get at 100% and average number of roots were 5.21. Transferred to pot mixture containing sphagna/yellow subsoil (1:2) after hardening, and the survival rate of plantlets could reached to 98.21%.
