摘要
目的:建立杨梅中二种主要黄酮类成分的同时含量测定方法。方法:采用Cosmosil-5C18-PAQ色谱柱(250mm×4.6mm,5μm);流动相为乙腈–0.1%乙酸溶液梯度洗脱;流速1.0m L/min;二极管阵列检测器,检测波长520nm(1)和360nm(2),同时测定杨梅中矢车菊素-3-葡萄糖苷(1)、杨梅苷(2)的含量。结果:二种黄酮类成分色谱分离良好,线性范围依次为20.08~200.8g/m L(r=0.9996,1)、3.984~39.84 g/m L(r=0.9995,2),加样回收率依次为97.5%、96.2%。结论 :该测定方法快速准确,可为杨梅的质量分析方法提供参考。
Objective To establish an HPLC method for the simultaneous determination of two fiavonoids in the fruits of Myrico rubra. Methods The chromatographic separation was performed on a Cosmosil - 5C18 - PAQ column (250 mm 4. 6 mm, 5 μm) with a gradient elution of acetonitrile and 0. 1% acetic acid solution, at a flow rate of 1.0 mL/min, and detected at both 520 nm (for cyanidin -3 -gluco- side, 1 ) and 360 nm (for myricetrin 2) using diode array detector. Results The two compounds were separated with satisfaction, with lineari- ty ranges between 20. 08 -200. 8 g/mL (r =0. 9996, 1) and 3. 984 -39. 84 g/mL (r =0. 9995, 2), and average recovery rates to be 97. 5% and 96. 2%. Conclusion The method is accurate and selective, which could be applied for the quality analysis of Myrico rubra.
