摘要
目的:以人参与西洋参为例,研究中药材不同等级规格商品中化学成分含量等信息,为中药材商品等级评价研究提供一种新方法。方法:采用RP-HPLC法测定人参中人参皂苷类成分或文献报道的西洋参的人参皂苷类数据,用SPSS 23.0软件分析商品等级与化学指标之间的相关关系,以人参或西洋参中与商品等级存在显著相关性的化学指标进行聚类分析。色谱条件:Alltech 100A氨基柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.2%磷酸(83∶17),流速1.0 mL·min^(-1),柱温35℃,检测波长203 nm。结果:以获得的17个人参皂苷类色谱峰面积数据或西洋参的8个人参皂苷含量数据的研究表明,人参或西洋参栽培时间、主根长度对化学成分的含量或含量比例具有较大影响,且栽培时间、主根长度与一些化学成分之间具有显著的相关性。聚类分析也表明,人参或西洋参中与商品等级存在显著相关性的化学指标能够明显提高商品等级之间的分类结果。这些与中药材商品等级相关的指标结合对照指纹图谱方法可以建立中药材商品的等级评价标准。结论:中药材商品规格等级标准适用于多年生的"道地药材"或按GAP规范生产的中药材商品规格的等级评价。
Objective:Taking ginseng and American ginseng as examples,the content of chemical components in different grades of Chinese medicinal materials was studied.A new method was provided for the evaluation of the grade of Chinese medicinal materials.Methods:The reported ginsenosides in Panax ginseng and Panax quinquefolium were determined by RP-HPLC.The correlation between the grade and the chemical variables was analyzed by cluster analysis of the chemical indexes with significant positive correlation,using SPSS 23 software.The Alltech Amino 100 A column(250 mm×4.6 mm,5 μm)was used,and the mobile phase was acetonitrile-0.2% phosphoric acid(83∶17).The flow rate was 1.0 mL·min^(-1) and the column temperature was controlled at 35 ℃.The detection wavelength was 203 nm.Results:The peak areas of 17 ginsenosides or the contents of 8 ginsenosides showed that the growth time and length of the main root of Panax ginseng and Panax quinquefolium had a great influence on the content or proportion of chemical components,and there was a correlation between them and the chemical components.Cluster analysis also showed that there was a significant positive correlation between the chemical indexes in the ginseng or American ginseng and the commodity grade,which could obviously improve the classification results between the grades.Combined with the reference fingerprints,the above indexes could establish the grade of Chinese medicinal materials.Conclusion:The grade standard of Chinese medicinal materials was applicable in the grade evaluation of the specifications of perennial "genuine medicinal materials" or Chinese medicinal materials produced according to the GAP.
